Identification of multiple constituents in shuganjieyu capsule and rat plasma after oral administration by ultra-performance liquid chromatography coupled with electrospray ionization and ion trap mass spectrometry

Abstract: Shuganjieyu (SGJY) capsule is a classical formula widely used in Chinese clinical application. In this paper, an ultra-performance liquid chromatography coupled with electrospray ionization and ion trap mass spectrometry has been established to separate and identify the chemical constituents of SGJY and the multiple constituents of SGJY in rats. The chromatographic separation was performed on a C18 RRHD column (150 × 2.1 mm, 1.8 μm), while 0.1% formic acid–water and 0.1% formic acid–acetonitrile was used as mo… Show more

“…The mass spectrum of this compound also contained an intense fragment ion with m/z = 193.0387 which probably corresponded to the neutral loss of dehydrated caffeic acid residue (162 Da, C 9 H 6 O 3 ). A comparison with published data [45,46] brought the conclusion that Compound 1 is an isomer of caffeoylquinic acid. Based on the analysis of the mass spectrum, Compound 2 with a retention time of 1.97 min was identified as quinic acid: [M + H]+ m/z 193.0428 and [M + Na]+ 215.0274 (calculated value for C7H13O6 − 193.0712; calculated value for C 7 H 12 O 6 Na − 215.0532).…”

“…In fruits and vegetables, flavonols generally present themselves as glycosides based on aglycons, such as kaempferol, quercetin, myricetin, isorhamnetin, and rhamnetin (Terahara, 2015). Isorhamnetin, the precursor (Xiao et al, 2018).…”

“…The MS/MS spectra confirmed the identity of quercetin 3- O -glucosyl-xyloside, which showed a product ion at m / z 265 [M–H−glucose−xylose, loss of 330] [ 50 ]. Similarly, the MS/MS spectra of quercetin 3- O -arabinoside also showed the product ion [M–H] − at m / z 301 [M–H−arabinoside, loss of 132] [ 71 ]. Both of these flavonoid-3- O -glycosides (compounds 43 and 46 ) have a flavonoid moiety that is O -glycosidically linked to the carbohydrate moiety at the C3-position.…”
