Identification of Long-Term Repopulating Potential of Human Cord Blood-Derived CD34−flt3− Severe Combined Immunodeficiency-Repopulating Cells by Intra-Bone Marrow Injection

Abstract: Recently, we have identified human cord blood (CB)-derived CD34-negative (CD34 ؊ ) severe combined immunodeficiency (SCID)-repopulating cells (SRCs STEM CELLS 2007;25:1348 -1355 Disclosure of potential conflicts of interest is found at the end of this article.

“…It has been shown that MSCs can reconstitute the BM microenvironment after direct injection into the BM. 31,32 In addition, MSCs have been found in BM after systemic infusion in an animal model and can enhance the engraftment of HSCs. [33][34][35] These results suggest that MSCs can home to BM and support hematopoiesis.…”

“…The CB-derived lineage-negative (Lin À ) mononuclear cells were separated using a StemSep device (StemCell Technologies, Vancouver, BC, Canada), as reported earlier. [7][8][9] Purification of Lin À CD34 þ CD38 þ , Lin À CD34 þ CD38 À , and Lin À CD34 À cells…”

“…[7][8][9]13 In this study, purified 2.5 Â 10 3 to 2 Â 10 4 CB-derived Lin À CD34 þ CD38 þ , 20-4 Â 10 2 CBderived Lin À CD34 À CD38 À cells, or 2-7 Â 10 4 CB-derived Lin À CD34 À cells were transplanted by IBMI 7-9 into sublethally irradiated (250 cGy using a 137 Cs-g irradiator) 8-to 10-week-old NOD/SCID or NOG mice. The repopulation of human hematopoietic cells in murine BMs was serially analyzed by the aspiration method.…”
