Identification of informative markers of agronomic traits in different ecotypes of sand plantain (Plantago psyllium)

Abstract: To investigate the relationships between agronomic traits and molecular markers in sand plantain (Plantago psyllium), a set of 17 ecotypes collected in Iran were evaluated. Twelve ISSR primers were tested through multiple regression analysis to identify the marker-trait association between ISSR data and agronomic traits. Finally, the 12 primers amplified 129 fragments (loci) among ecotypes: 91 fragments (70.54%) of the 129 fragments showed polymorphism with an average of 7.58% per primer and the rate of this p… Show more

“…Out of a total of 126 amplified DNA bands, up to 66 polymorphic DNA bands accounted for 66.2% with an average of 6.6 polymorphic bands appearing on each primer. This finding is consistent with previous study of Rahimi and colleagues when using the same ISSR primer set and found the polymorphism of 69.91% in different ecotypes of sand plantain (Rahimi et al 2017). This data also demonstrates that the primers used in the study are capable of amplifying in highly genetically distinct regions between gac samples.…”

“…The mean PIC obtained for the ISSR primers (=0. 2727) in the present study was lower than the mean PIC (=0.36) estimated for 12 ISSR primers in P. psyllium in the Rahimi et al[36] study. This result shows the ISSR primers tested in our study were informative with respect to discerning genetic diversity of the eight Plantago species.…”

“…Presently, analysis of 25 ISSR primers resulted in above 80% polymorphism detected in the 31 Plantago accessions. In other studies with Plantago 19.35% (Kaswan et al[4] and 70.54%[36] polymorphisms were identified for ISSR primers tested in P. ovata and P. psyllium, respectively. Analysis of PIC analysis provides an estimate of the discriminating power of a marker.…”

“…The high value of the MI for ISSRs is the result of highlights the distinctive nature of these markers. The higher the MI, the better method is used (Rahimi et al, 2018).…”
