Identification of ypqP as a New Bacillus subtilis Biofilm Determinant That Mediates the Protection of Staphylococcus aureus against Antimicrobial Agents in Mixed-Species Communities

Abstract: In most habitats, microbial life is organized in biofilms, three-dimensional edifices sustained by extracellular polymeric substances that enable bacteria to resist harsh and changing environments. Under multispecies conditions, bacteria can benefit from the polymers produced by other species ("public goods"), thus improving their survival under toxic conditions. A recent study showed that a Bacillus subtilis hospital isolate (NDmed) was able to protect Staphylococcus aureus from biocide action in multispecies… Show more

“…Normally, the excision of the SPβ prophage from the B. subtilis chromosome takes place before sporulation and allows reconstitution of the spsM gene involved in spore polysaccharide biosynthesis42. Sanchez-Vizuete et al 43. demonstrated that removal of SPβ from the chromosome permanently restores spsM , resulting in increased biofilm thickness.…”

“…Although the genetic construction of the strains can be difficult and time consuming, this approach has the unique advantage to be compatible with real-time 4-D (x-y-z-time) biofilm imaging (Klausen et al, 2003a). In the context of multispecies biofilms, this has allowed the analysis of interspecies competition and interference in-between species (Bridier et al, 2014), and identifying key molecular determinants involved in biofilm formation (Klausen et al, 2003a;Sanchez-Vizuete et al, 2015). Limitations of biofilm analysis by CLSM include: (i) interference of local physico-chemical properties of the biofilm with fluorescence probes and (ii) natural auto-fluorescence of the sample hiding the signal of interest.…”

“…Maximum biofilm thickness (in micrometers) was measured directly from xyz stacks. Biovolumes (in cubic micrometers) were calculated by binarizing images with a java script executed by ICY software as described previously (21). The biovolume was then defined as the overall volume of cells in the observation field.…”
