Identification of ‘Candidatus phytoplasma ziziphi’ associated with persimmon (Diospyros kaki Thunb.) fasciation in China

Abstract: Persimmon (Diospyros kaki Thunb.) trees, with fasciation symptoms (PF), were observed in an orchard located in suburban Tai'an, Shandong Province, China. A specific fragment of the phytoplasma 16S rRNA gene, approximately 1.2 kb in length, was amplified from two symptomatic plants via nested polymerase chain reaction, while no fragment was obtained from healthy controls. The two samples (PF1 and PF2) resulted with 99.5% nucleotide sequence identity. Phylogenetic and restriction fragment length polymorphism (RF… Show more

“…The 'Ca. P. ziziphi' species includes strains from a homogenous ecological lineage (Wang et al, 2018a), although isolates with less than 99.7% variation on their 16S rRNA sequence have been associated to similar stem fasciation of persimmon trees in China (Wang et al, 2015a). Also, Indian16SrV-B isolates infecting peach differ from 'Ca.…”

“…The ‘Lizao’ and ‘Dongzao’ jujube varieties appear to be susceptible to phytoplasma infection (Tian et al., 2000; Peng et al., ). Production of phytoplasma‐free plants of Chinese jujube can be achieved by cryopreservation (Wang et al., ,)…”

“…Flat stem often manifests as thickened and banded stems, disordered leaves, and abnormal in orescence development, affecting plants' normal growth and development. At present, the occurrence of at-stem disease has been reported in multiple countries and occurred on a variety of broad-leaved trees, including Sophora japonica (Liu et al 2022), Amorpha fruticose (Huang et al 2023), and Persimmon (Wang et al 2017), etc. It also affects herbaceous plants such as Corn (Duduk and Bertaccini 2006), Cucumber (Wang et al 2022), Arabidopsis thaliana (Bressan and Purcell 2005; Cettul and Firrao 2011), etc.…”
