Identification of cfxR, an activator gene of autotrophic CO2 fixation in Alcaligenes eutrophus

Abstract: A regulatory gene, cfxR, involved in the carbon dioxide assimilation of Alcaligenes eutrophus H16 has been characterized through the analysis of mutants. The function of cfxR is required for the expression of two cfx operons that comprise structural genes encoding Calvin cycle enzymes. CfxR (34.8 kDa) corresponds with an open reading frame of 954 bp, with a translational initiation codon 167 bp upstream of the chromosomal cfx operon. The cfx operon and cfxR are transcribed divergently. The N-terminal sequence … Show more

“…3B, lanes 2 and 3). These values agreed totally with those (33, (40); the X. flavus orfD sequence was partially determined, and the 150-amino-acid portion of the amino terminus was deduced (20). These proteins shared a high level of sequence identity (37.9 to 46.3%) for all regions of the molecule (Fig.…”

“…In addition to this high level of homology, T. ferrooxidans RbcR possesses the above-described common features, although some The function of this family of proteins requires binding to a specific target sequence(s); i.e., the operator and promoter regions of the genes they regulate. CfxR of A. eutrophus bound to the intergenic region between the cfxL and cfxR genes (40). As expected, RbcR, present in crude extracts from E. coli carrying cloned T. ferrooxidans rbcR, bound specifically to the intergenic sequence between rbcR and rbcLl (Fig.…”

“…A regulatory gene cbbR, encoding a transcriptional activator, belongs to the chromosomal locus. A plasmid-borne copy of cbbR is present but defective, so the chromosomal copy controls the coordinate expression of both operons 17 . Two substrates supply energy for autotrophic CO 2 fixation.…”

“…Adjacent to the cbbR gene is an ORF oriented divergently from cbbR; from comparison to a recently determined nucleotide sequence of a cbb gene from A. eutrophus (54), this ORF was identified as the pentose-5-phosphate 3-epimerase gene (cbbE). The translated sequence of the ORF is 53% identical to cbbE fromA.…”
