Identification of human tear fluid biomarkers in vernal keratoconjunctivitis using iTRAQ quantitative proteomics

Abstract: LC MALDI MS and isobaric tag for relative and absolute quantitation technique may be useful in the quantitative and qualitative characterization of the peptidoma of human tears. These techniques may identify target proteins to be used in the diagnosis and management of VKC and other inflammatory ocular surface conditions.

“…For the tear fluid proteome, up to around 1500 proteins have been identified in deep proteomic studies 7. Leonardi et al5 found 78 different proteins in vernal keratoconjunctivitis by iTRAQ quantitative proteomics which is a similar number of proteins found in the present study. Souza et al4 identified a large number of protease and protease inhibitors in tear fluid of one healthy subjects which was similar to findings we made in nasal mucus proteome.…”

“…Tear composition reflects the physiological condition of underlying tissue but can be altered in disease related to the eye or other organs and systemic diseases such as diabetes 2. Several tear proteomic studies for diseases other than allergic rhinoconjunctivitis exist proofing the feasibility of proteomic approaches to tear fluid 3, 4, 5, 6…”

“…Chemical labeling with iTRAQ or TMT makes possible the labeling of up to 8 or 10 different samples with an isobaric label [38][39][40][41][42][43]. The label is constructed in that way that the m/z values of the peptides originating from different samples (precursor ions) are the same but during fragmentation the labeling group is released and the rest of the peptide can be used for MS/ MS based protein identification.…”

“…Open to the outside environment, the eye is widely exposed to airborne allergens. In certain cases, investigations for ocular allergy require in situ collection of samples or in vivo challenge of the ocular surface by instilling allergens . Isolated from the body, the eye may react independently to allergens.…”

“…In VKC, which is considered as an etiopathogenic model of allergic conjunctivitis, conjunctival scrapings found massive infiltration of T cells and especially eosinophils. Multiple cytokines are released in particular TH2 cells induced cytokines . Specific IgE‐mediated hypersensitivity was evidenced in at least 50% of cases .…”
