Identification of genetically high risk individuals to lung cancer by DNA polymorphisms of the cytochrome P45 0IA1 gene

Abstract: A good correlation was observed between enhanced lung cancer risk and restriction fragment length polymorphisms (RFLPs) of the P450IAl gene with the restriction enzyme. MspI. Genotype frequencies of 0.49 for the predominant homozygote, 0.40 for the heterozygote, and 0.11 for the homozygous rare allele were observed in a healthy population. Among lung cancer patients, the frequency of homozygous rare allele of P450IAl gene was found to be about 3-fold higher than that among healthy population, and this differen… Show more

“…The CYP1A1 gene also plays an important role in the metabolism of estrogen in vivo, by catalyzing 2-hydroxylation. The 2-hydroxylation products (2-OH estradiol catechol and 2-OH estrone catechol) lack estrogenic activities (Kawajiri et al 1990). Furthermore, the 2-hydroxy catechol metabolites can be converted by Omethylation into 2-methoxy derivatives, which have been shown to possess anti-proliferative and anti-angiogenic properties (Cascorbi et al 1996;Crofts et al 1993).…”

“…Thus, the idea was considered to be a matter of controversy (13). Afterwards, the eŠects of genetic polymorphism of CYP1A1 on the lung cancer risk were demonstrated in Japanese by Kawajiri and his associates (14). This idea seemed to be reasona- Among many forms of cytochrome P450, CYP2A6 is known to be responsible for the metabolism of drugs including coumarin (18,19), nicotine (20), tegafur (21,22), fadrozole (23), methoxy‰urane (24) and valproic acid (25).…”
