Identification of genes specifically expressed in maternal and filial tissues of barley caryopses: a cDNA array analysis

Abstract: Developing seeds consist of genetically distinct maternal and filial tissues, whose interactions during development are largely unknown. To better understand the molecular physiology of developing seed tissues in barley, we created a high-density cDNA macroarray bearing 711 cDNA fragments from 691 clones representing at least 620 unique genes mainly derived from a cDNA library constructed with mRNA from the early stages of caryopsis development. This array has been used to compare gene expression patterns in m… Show more

“…Array hybridizations were performed at least twice per each time point using different biological replicates of plant material. The synthesis of 33 P-labeled cDNA was performed as described by Sreenivasulu et al [ 30 ]. Hybridizations were carried out overnight at 65°C in Church buffer (250 mM sodium phosphate, pH 7.2, 7% (w/v) SDS, 1% (w/v) BSA, 1 mM EDTA).…”

“…The key to this system is that individual resistant and infected epidermal cells of susceptible barley can be distinguished easily by microscopic observation of B. graminis hordeichallenged barley leaves at 18 hai (55). Subsequently, the contents of these and noninocu-lated control cells were collected for mRNA extraction, amplification, and hybridization to the IPK (Gatersleben) barley PGRC1 10k cDNA array (56,129). Hierarchical clustering and multidimensional scaling (31) were used to visualize the relationships between samples.…”

“…The overexpressing HvGAMYB transgenic lines show reduced anther size with a male sterility phenotype [75]. Our laboratory has recently characterized a new protein called Jekyll, which is preferentially expressed in barley grain nucellar projection tissue [76]. Its downregulation decelerates autolysis of nurse tissue.…”
