Antioxidant and Hepatoprotective Activity of Abelmoschus Manihot L. Medik Leaf Fraction Against Ccl4-Induced Liver Damage in Rats

Objective: The objective of this research was to determine the antioxidant and hepatoprotective activity of Abelmoschus manihot L. Medik (AMLM) leaves in carbon tetrachloride-induced liver damage in rats (Rattus norvegicus). Methods: Samples of dried leaf were macerated using 96% ethanol solvent to obtain a viscous extract. Fractionation was then performed using ethyl acetate and n-butanol solvent. The antioxidant activity test of each fraction was done by 1,1-diphenyl-2-picrylhydrazyl assay. For the hepatoprotective activity test, the fractions were suspended in Tween 20 0.4% solution with concentrations of 10 mg/mL and 20 mg/mL. Rats were divided into 6 treatment groups consisting of five rats/group. Group I (positive control) was given an oral suspension of Curcuma® tablet, Group II (negative control) was given Tween 20 0.4% solution, and Groups III and IV (ethyl acetate fraction) and V and VI (n-butanol fraction) were given fraction suspension. On the 1st–7th days, all groups were given the solution treatment, and on the 8th day, all groups were injected with CCl4 intraperitoneally and the treatment was continued until the 11th day. On the 12th day, the blood of rats was taken and then measured the serum glutamic oxaloacetic transaminase (SGOT) and serum glutamic pyruvic transaminase (SGPT). Results: The results of the antioxidant test showed that ethyl acetate and n-butanol fraction, respectively, had IC50 of 89.99 and 114.56 ppm.Measurement of SGOT showed the result for Groups I–VI, respectively, of 160±63.62, 260.53±18.98, 154.16±52.78, 177.43±13.70, 120.07±34.80, and 105.23±40.49 IU/L. Measurement of SGPT showed the result for Groups I–VI, respectively, of 101.87±29.24, 108.1±9.04, 57.73±49.05, 106.07±26.45, 66.9±20.05, and 146.63±84.89 UI/L. Conclusion: The results of this research indicated that the ethyl acetate and n-butanol fraction of AMLM leaves have the antioxidant andhepatoprotective activity.

Obtaining high-added value compounds from agricultural waste receives increasing attention, as it can both improve resource utilization efficiency and reduce waste generation. In this study, polysaccharides are extracted from the discarded roots of Abelmoschus manihot (L.) by the high-efficiency ultrasound-assisted extraction (UAE). The optimized condition was determined as solid–liquid ratio SL ratio = 1:20, temperature T = 30 °C and time T = 40 min, achieving an extraction yield of 13.41%. Composition analysis revealed that glucose (Glc, 44.65%), rhamnose (Rha, 26.30%), galacturonic acid (GalA, 12.50%) and galactose (Gal, 9.86%) are the major monosaccharides of the extract. The extract showed a low degree of esterification (DE) value of 40.95%, and its Fourier-transform infrared (FT−IR) spectrum exhibited several characteristic peaks of polysaccharides. Inspired by the wide cosmetic applications of polysaccharides, the skincare effect of the extract was evaluated via the moisture retention, total phenolic content (TPC) quantification, 2,2-Diphenyl-1-picrylhydrazyl (DPPH)-free radical scavenging activity, anti-hyaluronidase and anti-elastase activity experiments. The extract solutions demonstrated a 48 h moisture retention rate of 10.75%, which is superior to that of commercially available moisturizer hyaluronic acid (HA). Moreover, both the TPC value of 16.16 mg GAE/g (dw) and DPPH-free radical scavenging activity of 89.20% at the concentration of 2 mg/mL indicated the strong anti-oxidant properties of the extract. Furthermore, the anti-hyaluronidase activity and moderate anti-elastase activity were determined as 72.16% and 42.02%, respectively. In general, in vitro skincare effect experiments suggest moisturizing, anti-oxidant, anti-radical and anti-aging activities of the A. manihot root extract, indicating its potential applications in the cosmetic industry.

Medicinal plants are of great significance and global importance as antioxidants and antibacterial agents. In the present investigation the chloroform leaf extract of Abelmoschus manihot was screened for its phytochemical constituents, antioxidant and antibacterial properties. Flavonoids, phenolics, tannins and vitamins were the phytochemical constituents present. GC-MS analysis revealed ten major phytocompounds. In vitro antioxidant assay via DPPH exhibited an IC50 value of 234.7±37.53mg/mL, and its activity could be ascribed due to the presence of the phytocompound α-tocopherol. The antimicrobial activity represented by the zone of inhibition via disc diffusion method, observed no activity against the fungal strains, Aspergillus niger and Candida albicans. Staphylococcus aureus and Escherichia coli exhibited 12 to 14mm zone of inhibition at concentrations 100-500µg, which can be attributed due to the presence of flavonoids. The above results suggests that antioxidant and antibacterial activity exhibited by the chloroform leaf extract of this plant was attributed to the presence of the phytochemicals, especially phenolics and flavonoids. Further in depth studies on the phytochemical constituents of this chloroform leaf extract is recommended, as this extract could serve as an additional implement to natural antioxidant and antibacterial agents.
