Identification of Five Candidate Lung Cancer Biomarkers by Proteomics Analysis of Conditioned Media of Four Lung Cancer Cell Lines

Abstract: Detection of lung cancer at an early stage is necessary for successful therapy and improved survival rates. We performed a bottom-up proteomics analysis using a twodimensional LC-MS/MS strategy on the conditioned media of four lung cancer cell lines of different histological backgrounds (non-small cell lung cancer: H23 (adenocarcinoma), H520 (squamous cell carcinoma), and H460 (large cell carcinoma); small cell lung cancer: H1688) to identify secreted or membrane-bound proteins that could be useful as novel lu… Show more

“…The mechanisms underlying the distinct molecular features observed in ADC-TAFs and SCC-TAFs remain unknown. However, they could be associated with differences in the secretome of cancer cells from these two subtypes (44). Unlike stiff substrata, no marked differences in absolute (not normalized) cell density values were observed in 1 kPa gels between ADC-TAFs and SCC-TAFs ( Supplementary Fig.…”

“…PTX3 protein or TNF inducible gene 14 encoded by PTX3 gene in humans is secreted by different cell types such as endothelial cells, mononuclear phagocytes, dendritic cells and fibroblasts upon stimulation with primary inflammatory cytokines and toll like receptor engagement. Planque et al [152] found that serum levels of PTX3 were much higher in lung cancer patients (median = 4.91 ng/ml) compared to healthy individuals (median = 1.52 ng/ml). The diagnostic sensitivity of pentraxin at 100% specificity was found to be 68%.…”

“…Any of these play key roles in a variety of diseases, including cancer. Aberrant expression of ADAM17 has been shown in various malignancies (17)(18)(19). However, reports on the expression and function of ADAM17 in prostate cancer are limited.…”
