Identification of estrogenic and antiestrogenic activities of respirable diesel exhaust particles by bioassay-directed fractionation

Abstract: Bioassay-directed fractionation was performed to identify causative chemical groups of DEPs with estrogenic and antiestrogenic activities. Bioassay-directed fractionation consists of a cell bioassay (E-SCREEN) in conjunction with acid-base partitioning (F1 and F2) and silica gel column fractionation of neutral fractions (F3-F7). Crude extract (CE) of DEPs in dichloromethane (DCM) exhibited both estrogenic and antiestrogenic activity. Estrogenic activity of CE and some fractions (F1, F2, F3, F5 and F6) was indu… Show more

“…In regards to this type of analysis, sample screening was performed by using a yeast estrogen screen (YES) assay for total estrogenic activity; then, the analysis for specific chemical species was conducted by using LC-MS/MS [85,111]. Another application method was bioassay-directed fractionation, which consists of a cell bioassay (E-SCREEN), in conjunction with acid-base partitioning (F1 and F2) and the silica gel column fractionation of neutral fractions (F3-F7) [112]. In addition, the recombinant yeast assay (RYA), combined with chemical identification by LC-MS, was used [113].…”

“…contained in PM, especially from diesel exhaust [12,13], are described in the literature as endocrine disruptors with either estrogenic, antiestrogenic or anti-androgenic activity [14,[77][78][79][80][81]. Kizu et al described anti-androgenic activity of certain particle compounds from diesel exhaust in human PC3/ AR cells derived from prostate cancer tumors [15], while Okamura et al reported anti-estrogenic activity of similar compounds in human MCF-7 cells from breast cancer tumors [14].…”

“…These hormone disruptions may be induced by air pollutants, notably PAHs, binding to estrogen receptors [78,79,82,83] or androgen receptors [15,84] with an agonistic or antagonistic effect. They may also be the result of activation of AhR pathway.…”

“…Substances that cross the placenta and/or are secreted into milk might reach the brains of embryos or neonates while the blood-brain barrier is immature (Dalgaard et al 2001;Oh et al 2008). Thus, the possibility that exposure to EDC influences the central nervous system (CNS) or leads to neurodegenerative disorders in adulthood cannot be ruled out.…”
