Identification of CRMP4 as a Convergent Regulator of Axon Outgrowth Inhibition

Abstract: Myelin-associated inhibitors (MAIs) and chondroitin sulfate proteoglycans (CSPGs) contribute to failed regeneration after neuronal injury. MAIs and CSPGs stimulate intracellular signals including the activation of RhoA and Rho kinase to block axonal extension through targeted modifications to the cytoskeleton. RhoA and ROCK are promising targets for therapeutic intervention to promote CNS repair; however, their ubiquitous expression will limit the specificity of drugs targeted to these molecules. We have ident… Show more

“…Constructs-The HSV-CRMP4S-V5 construct was described previously (28). CRMP4S, CRMP4⌬N300 (amino acids 300 -570), CRMP4⌬C50 (amino acids 1-520), CRMP4⌬C100 (amino acids 1-470), and CRMP4⌬470 -520 (deletion of amino acids 470 -520) DNAs were amplified by PCR from the pcDNA3.1/V5-His-CRMP4S construct and subcloned into the pHSV-PUC-IRES-GFP vector using SalI and XbaI restriction sites.…”

“…Additionally, seven genes were identified that were categorized by the gene ontology biological process GO:0007399 (nervous system development) (P Ͻ 0.04). Three of these genes encode components of signal transduction pathways that regulate the movement and/or development of neurons within the embryonic brain: chemokine (C-X-C motif) receptor 4 (Cxcr4), glypican 4 (Gpc4), and dihydropyrimidinaselike 4 (Dpysl4) (30)(31)(32)(33)(34)(35). For example, Cxcr4 regulates the migration of embryonic neurons into the cerebellum (35).…”
