Identification of Cows Treated with Recombinant Bovine Somatotropin

Abstract: A method for the specific detection and quantification of recombinant bovine somatotropin (rbST) in bovine blood has been validated according to criteria described in the EU Commission Decision 2002/657/EC. The method is based on a thorough purification procedure followed with the detection by LC-ESI-MS/MS of the tryptic N-terminal peptide specific of the rbST. The recombinant equine somatotropin (reST) is used as internal standard. Performance of the method was assessed based on specificity, linearity, truene… Show more

“…For the three best-performing prediction models IA, AO and IAO, false-positive rates ranged from 10.6% to 14.7%, which was quite acceptable, since samples screened positive must be analysed by a subsequent confirmatory analysis method according to Commission Decision 2002/657/EC anyway [23]. The confirmation method is based on the detection of an N-terminal peptide of somatotropin, which has a different terminal amino acid in the recombinant form of the hormone [26].…”

“…Note that in practice, a whole stable and not an individual cow is treated with rbST, thus increasing the chance of detecting rbST use. And third, for final confirmation of rbST itself in serum samples of suspect individual cows, a highly sensitive mass spectrometric confirmatory method, which fulfils the 2002/657/EC confirmatory method requirements, is to be used [26].…”

“…Direct detection of rbST, however, is problematic due to its short half‐life in blood, the similarity with the endogenous hormone (bST, also called growth hormone), the low concentrations of bST and rbST in serum, and strong fluctuations therein. Preliminary results regarding the development of a confirmation method for rbST detection based on liquid chromatography‐mass spectrometry (LC‐MS) show potential . But that method is rather laborious, time‐consuming, and requires highly trained personnel and expensive equipment.…”

“…In contrast to MS‐based sports drug testing for hGH, several MS methodologies for identification of rGH in bovine and equine species have been developed (Blokland et al, 2003, ; Le Breton et al, 2008, ; Bailly‐Chouriberry et al, ). Identification of rGH in these species is possible by selective measurement of N‐terminal tryptic peptides that contain small amino acid substitutions (Fig.…”
