Identification of circular dorsal ruffles as signal platforms for the AKT pathway in glomerular podocytes

Abstract: Circular dorsal ruffles (CDRs) are rounded membrane ruffles induced by growth factors to function as precursors of the large‐scale endocytosis called macropinocytosis. In addition to their role in cellular uptake, recent research using cell line systems has shown that CDRs/macropinocytosis regulate the canonical AKT–mTORC1 growth factor signaling pathway. However, as CDRs have not been observed in tissues, their physiological relevance has remained unclear. Here, utilizing ultrahigh‐resolution scanning electro… Show more

“…The initial and critical step in macropinocytosis involves membrane ruffling or blebbing, which appears to be regulated by various PIPs [25,27]. Inhibitors of phosphoinositide 3-kinases (PI3Ks), responsible for generating PI (3,4,5)P3 from PI(4,5)P 2 , can impair macropinosome formation [28].…”

“…Closure of ruffles is completed by fusion of ruffles, resulting in the formation of large vesicles (>0.2 μm in diameter) called macropinosomes (Conner & Schmid, 2003; Quinn et al, 2021; Swanson, 2008). Significant progress has been made in understanding the responsible genes and molecular mechanisms governing MPC induction and macropinosome maturation in various cell types (Fennell et al, 2015; Freeman et al, 2020; Hua et al, 2023; Kawai et al, 2021; Maekawa et al, 2014; Mylvaganam et al, 2021; Ramirez et al, 2019; Stow et al, 2020; Sun et al, 2022; Yoshida et al, 2018; Zeziulia et al, 2022).…”
