Identification of circadian rhythms in Nannochloropsis species using bioluminescence reporter lines

Abstract: Utilization of microalgae has been hampered by limited tools for creating loss-of-function mutants. Furthermore, modified strains for deployment into the field must be free of antibiotic resistance genes and face fewer regulatory hurdles if they are transgene free. The oleaginous microalga, Nannochloropsis oceanica CCMP1779, is an emerging model for microalgal lipid metabolism. We present a one-vector episomal CRISPR/Cas9 system for N. oceanica that enables the generation of marker-free mutant lines. The CEN/A… Show more

“…Cas9-mediated target gene disruption was conducted using an episomal CRISPR system 51 . The sgRNAs were designed to meet the following criteria: (1) uniqueness of target sequence and PAM (final 12 bp + NGG) in genome, (2) location within the first 300 bp of the gene, and (3) target sequence present in both genome and mRNA sequence (to avoid targeting introns).…”

“…Robust diel rhythms in growth, gene expression, pigment synthesis, phototactic movements, and bioluminescence have been observed in a variety of phytoplanktonic organisms, including diatoms (710). Diatoms represent the most species-rich group of algae in the ocean and populate a wide range of aquatic environments (11).…”

“…This system efficiently generates targeted mutations and allows the loss of episomal DNA after removing the selection pressure, resulting in marker-free non-transgenic engineered lines. In the study reported by Poliner et al, 2019 [16], the nitrate reductase gene (NR) has been disrupted and subsequently, the CRISPR episome has been successfully removed to generate non-transgenic marker-free nitrate reductase knock-out lines (NR-KO).…”

“…Despite the simplicity of microalgae cells, few of the CRISPR-Cas-mediated transformation and genetic editing have been successful ( [14][15][16]). Through all the attempts and trials to edit target genes in microalgae, many pitfalls have been encountered harming and slowing down the research in this field.…”
