Identification of central amygdala and trigeminal motor nucleus connectivity in humans: An ultra‐high field diffusion MRI study

Abstract: The neuroanatomical circuitry of jaw muscles has been mostly explored in non‐human animals. A recent rodent study revealed a novel circuit from the central amygdala (CeA) to the trigeminal motor nucleus (5M), which controls biting attacks. This circuit has yet to be delineated in humans. Ultra‐high diffusion‐weighted imaging data from the Human Connectome Project (HCP) allow in vivo delineation of circuits identified in other species—for example, the CeA–5M pathway—in humans. We hypothesized that the CeA–5M ci… Show more

“…Studies in mice have revealed that a direct projection from CeA to trigeminal motor nucleus is highly activated during hunting of prey and that it promotes strong biting attacks (43). Of note, the same pathway has recently been identified in humans, and in higher distribution than BLA-Mo5 (Trigeminal Motor Nucleus) (44), further highlighting that the same association may also apply to humans and warranting further research in this field. “…There have been reports of direct projections from the central nucleus of the amygdala to the Me5, which are crucial for the perception and control of negative emotions (69). This type of projection has been connected to stronger biting attacks during hunting in animal studies and has recently been identified in humans as well (70,71).…”

“…Han et al 75 showed that a brain circuit connecting CeA to the trigeminal motor nucleus (5 M) controls the muscles of mastication during prey hunting in rodents. Of note, this circuit has also been recently delineated in human MRIs 76 . As CeA is a crucial regulatory hub for anxiety, it is possible that amygdala plays a key role in modulating jaw motor function and may be implicated in the neural pathways leading to bruxism‐like behaviours as a stress response.…”
