Identification of Calmodulin Isoform-specific Binding Peptides from a Phage-displayed Random 22-mer Peptide Library

Abstract: Plants express numerous calmodulin (CaM) isoforms that exhibit differential activation or inhibition of CaMdependent enzymes in vitro; however, their specificities toward target enzyme/protein binding are uncertain. A random peptide library displaying a 22-mer peptide on a bacteriophage surface was constructed to screen peptides that specifically bind to plant CaM isoforms (soybean calmodulin (ScaM)-1 and SCaM-4 were used in this study) in a Ca 2؉ -dependent manner. The deduced amino acid sequence analyses of … Show more

“…It is likely that the ability of NtCBK1 to be activated equally by all three tobacco CaMs is related to the fact that the CaM-binding domain of NtCBK1 possesses both 1-5-10 and 1-8-14 CaM binding motifs. It has been reported that different CaMs regulate their targets based on the types of the motifs present in their CaM-binding domains (56). We speculate that because NtCBK1 has both 1-5-10 and 1-8-14 CaM-binding domains, it can be activated by several different CaM isoforms.…”

“…For example, Arabidopsis has 11 CaM genes that encode for seven CaM isoforms. The different CaM isoforms differ in their ability to bind and activate known CaM-regulated enzymes in vitro (57,58). In this study, potato CaM PCM6 and bovine CaMs were used, and no significant differences were found in CaM binding and enzyme activation.…”

“…1B) (37). Moreover, the C-terminal region of the protein (amino acids 726 -845) contains a cluster of several putative CaM-binding domains, four Ca 2ϩ -dependent CaM dissociation domains (IQ motifs), and a Ca 2ϩ -dependent CaM-binding domain (38,39). These structural features suggest that this clone is a transcription factor whose activity can be modulated by CaM binding.…”
