Identification of angiogenic properties of insulin-like growth factor II in in vitro angiogenesis models

Abstract: Summary Insulin-like growth factor II (IGF-II), highly expressed in a number of human tumours, has been recently known to promote neovascularization in vivo. Yet, the detailed mechanism by which IGF-II induces angiogenesis has not been well defined. In the present study, we explored an angiogenic activity of IGF-II in in vitro angiogenesis model. Human umbilical vein endothelial cells (HUVECs) treated with IGF-II rapidly aligned and formed a capillary-like network on Matrigel. In chemotaxis assay, IGF-II remar… Show more

“…Numerous reports indicate that growth factors such as VEGF, bFGF, IGF-I and -II are involved in the angiogenic process in various tumours including breast cancer (Kandel et al, 1991;Grant et al, 1993;Heffelfinger et al, 1999;Neufeld et al, 1999;Lee et al, 2000;Yan et al, 2000). Our results demonstrate that the treatment of MDA-MB-435 oestrogen-independent breast cancers with bombesin/GRP antagonists RC-3095 and RC-3940-II significantly decreased the expression of mRNA for VEGF-A, b-FGF and IGF-II, as well as their protein levels.…”

“…Thus, there is a significant correlation between activated MMP-2 expression and VEGF expression, and similarly pro-MMP-9 expression tends to correlate with the increment of MVD (Kurizaki et al, 1998). IGF-II has also been described as an angiogenic factor that acts either directly (Volpert et al, 1996;Lee et al, 2000) or indirectly through an increase in VEGF production and the upregulation of the expression of proteolytic enzyme MMP-2 (Kim et al, 1998). Overactive MMPs contribute to an almost complete loss of the peritumoral basement membrane in most cancers including breast carcinoma (Lebeau et al, 1999).…”

“…The conditioned medium containing 5 g secreted proteins was analyzed by gelatin-and casein-based zymographies. 15 The digested area appeared clear on a blue background, indicating the location of matrix metalloproteinase (MMP) activity. To further verify MMP caseinolytic activity, the casein-containing gel was incubated with incubation buffer containing 10 mmol/L EDTA, an inhibitor of MMPs, to observe disappearance of the clear band.…”
