Identification of an overprinting gene in Merkel cell polyomavirus provides evolutionary insight into the birth of viral genes

Abstract: Many viruses use overprinting (alternate reading frame utilization) as a means to increase protein diversity in genomes severely constrained by size. However, the evolutionary steps that facilitate the de novo generation of a novel protein within an ancestral ORF have remained poorly characterized. Here, we describe the identification of an overprinting gene, expressed from an Alternate frame of the Large T Open reading frame (ALTO) in the early region of Merkel cell polyomavirus (MCPyV), the causative agent o… Show more

“…The most consistently detected region was at the 5′ end of LTAg exon 2 (also shared by ALTO and 57kT transcripts) (Supplementary Figure S2) (3,23,24), overlapping with a commonly targeted region for qPCR detection of MCPyV ( Figure 1). However, we and others have observed cases with high sTAg and low LTAg expression by qPCR (14,25).…”

“…RacPyV is closely related to MCPyV at both the genome and protein levels, and they both fall within a monophyletic clade of viruses that some have called almipolyomaviruses, which encode either an overprinting "ALTO" protein or a similarly overprinted middle tumor antigen (MT) protein (10). Almipolyomaviruses include PyVs that are causative for naturally occurring tumors.…”

“…The MCV tumor antigen is multiply spliced into the large tumor antigen (LT), the small tumor antigen, 57kT, and ALTO (alternate frame of the large T open reading frame) (12,13). Similar to other polyomaviruses, the multifunctional MCV LT protein is involved in a variety of processes, including viral genome replication and host cell cycle manipulation (14 -16).…”
