Identification of Alternative Splicing Events Regulated by an Arabidopsis Serine/Arginine-Like Protein, atSR45a, in Response to High-Light Stress using a Tiling Array

Abstract: We have demonstrated that an Arabidopsis serine/arginine rich-like protein, atSR45a, interacts with other splicing factors and its expression is markedly induced by high-light stress, suggesting the involvement of atSR45a in the regulation of stress-responsive alternative splicing. A whole-genome tiling array identified the alternative splicing of genes regulated by atSR45a by comparing gene expression profiles in wild-type and knockout atSR45a (KO-sr45a) plants under high-light stress. The expression levels o… Show more

“…Indeed, the expression of SR45 has been reported to show significant changes under sucrose and temperature stress [58], but not in response to ABA or other phytohormones [40,58]. Expression of SR45a is markedly induced by high light stress [59], while DNA microarray studies [60,61,62,63] and RNA-seq analysis of abiotic stress samples [64] have shown that SR45a mRNA abundance is also altered upon heat and water deprivation stresses. Nevertheless, SR45a expression in response to ABA was not investigated until this study.…”

“…RT-PCR showed that the expression of the selected gene in each mutant was completely abrogated (Figure S3). Some of these genes are known to participate in abiotic stress, such as EPR1 in light rhythm [37], NTL4 in ROS regulation during drought-induced senescence [87] and SR45a in alternative splicing under high light [88].…”

“…Expression of P5CDH was found to be regulated by gene silencing via the overlapping gene Sro5 on the complementary DNA strand, but so far, our analyses did not clearly demonstrate an altered regulation of P5CDH in the rsr1-1 mutant (Funck, 2001; Borsani et al, 2005). A recent study on transcriptome changes in a splicing factor mutant suggests that RSR1 / ESP1 itself may be regulated by alternative mRNA processing (Yoshimura et al, 2011). …”
