Identification of a β Subunit TM2 Residue Mediating Proton Modulation of GABA Type A Receptors

Abstract: GABA type A (GABA A ) receptors are functionally regulated by external protons in a manner dependent on the receptor subunit composition. Although H ϩ can regulate the open probability of single GABA ion channels, exactly what residues and receptor subunits are responsible for proton-induced modulation remain unknown. This study resolves this issue by using recombinant ␣1␤i subunit GABA A receptors expressed in human embryonic kidney cells. The potentiating effect of low external pH on GABA responses exhibited… Show more

“…For example, in the ␣1␤ Gly receptor activation by agonists is markedly affected by mutations in the ␣1 subunit, but not in the homologous ␤ mutant subunits (30). In the GABA A receptor, asymmetric contribution of subunits has been reported for permeability (31), effect of gain-of-function mutations (L9Ј and F6Ј) (32)(33)(34) and mutations that ablate pH modulation (35).…”

“…DEPC was used to specifically alkylate the imidazole group of histidines (22); this treatment resulted in the loss of the copper inhibition and a reduction in the P2X 4 ATP-evoked currents of the receptor. We are aware that there are controversial results reported in the literature with respect to the use of DEPC, because some authors have successfully modified histidine residues (23)(24)(25), whereas others have failed to detect significant changes (26,27). Considering that the rat P2X 4 receptor has only three histidine residues, it became challenging to establish which of these imidazole-bearing residues is responsible for the negative modulator role of copper, as well as the decreased channel activity.…”

“…Cell Culture and Transfection-Human embryonic kidney (HEK) cells (American Type Culture Collection CRL1573) were grown and transfected as previously documented (23). Plasmids of hGlyR cDNA clones were co-transfected in a ratio of 1:1 with enhanced green fluorescent protein (24).…”
