Identification of a TGF-β/SMAD/lnc-UTGF positive feedback loop and its role in hepatoma metastasis

Abstract: Aberrant activation of the TGF-β/SMAD signaling pathway is often observed in hepatocellular carcinoma (HCC). Whether lncRNA regulates the TGF-β/SMAD signaling remains largely unknown. Here, we identified an oncogenic lncRNA that was upregulated in HCC and was transcriptionally induced by TGF-β (named lnc-UTGF, lncRNA upregulated by TGF-β). Upon TGF-β stimulation, SMAD2/3 bound to the lnc-UTGF promoter and activated lnc-UTGF expression. In turn, the TGF-β/SMAD signaling was augmented by overexpressing lnc-UTGF,… Show more

“…Generally, its coding capacity is scarce, which was regarded as nonsense noises in transcripts 32,33 . However, several studies confirmed that lncRNAs have unique and important biological functions including gene regulation, RNA translocation, RNA stability modulation and translation regulation in tumourigenesis and development of tumours via various mechanisms 34–37 . Recent studies proved that RNA‐binding proteins participate in the lncRNA‐mediated gene regulation.…”

“…Activated by Smad2/3, Smad7 inhibits TGF-β/Smad signaling by binding to TGFβRI/II. [102] In the liver, hepatic sinusoidal endothelial cells and hepatic stellate cells are the main producers of TGF-β. TGF-β activity has been demonstrated to be key to HCC pathogenesis, including activation of tumor-associated fibroblasts (CAFs), tumor-associated macrophages, and other tumor-associated cells.…”

“…When TGF-β is present, it promotes SMAD2/ 3 binding to the lnc-UTGF promoter, which in turn stimulates transcription of lnc-UTGF, which then interacts with and stabilizes SMAD2/4 mRNA. HCC metastasis is facilitated by the loop because of its enhancement of TGF-β signaling (Wu et al, 2021).…”
