Identification of a set of miRNAs differentially expressed in transiently TIA-depleted HeLa cells by genome-wide profiling

Abstract: BackgroundT-cell intracellular antigen (TIA) proteins function as regulators of cell homeostasis. These proteins control gene expression globally at multiple levels in response to dynamic regulatory changes and environmental stresses. Herein we identified a micro(mi)RNA signature associated to transiently TIA-depleted HeLa cells and analyzed the potential role of miRNAs combining genome-wide analysis data on mRNA and miRNA profiles.ResultsUsing high-throughput miRNA expression profiling, transient depletion of… Show more

“…Hypoxia and ischemia in the tendon might have induced miR-498 which downregulated collagen type 1 expression but, additional studies are required to validate this aspect. In addition, miRNA-498 was found to be upregulated to control the metastasis of several cancer types including liver cancer, adenocarcinoma and retinoblastoma [45]. Also, increased expression of miR-498 has been correlated with the ECM damage associated with psoriasis [46].…”

“…The tissue sections were analyzed for the expression of Collagen I and Collagen III and MMP-2 and MMP-9 expression by immuno-double staining following our previously established standard protocols [13, 45, 50]. HIER buffer (Heat Induced Epitope Retrieval) was used for antigen retrieval at 95°C for 20 mins and the sections blocked in PBS containing 0.25% Triton X-100 and 5% horse serum for 2 hr at room temperature.…”

“…TIA proteins are known to target genes with relevant biological implications in apoptosis, inflammation, cell responses to stress, viral infections and oncogenesis [1], [2], [18], [20][24]. Further, these proteins seem to have an important role during embryogenesis.…”

“…People with a high level of miR-586 have relatively low expression levels of these genes, which leads to the high risk of aGVHD. Some target genes of miR-586 including SFPQ, KCTD12, FAM129A, TNRC6B, SLC12A2, and VPS13A were predicted using a bioinformatic tool [21]. However, the genes related to aGVHD need further elucidation.…”
