Identification of a Proline-Rich Sequence in the CD2 Cytoplasmic Domain Critical for Regulation of Integrin-Mediated Adhesion and Activation of Phosphoinositide 3-Kinase

Abstract: The CD2 molecule is one of several lymphocyte receptors that rapidly initiates signaling events regulating integrin-mediated cell adhesion. CD2 stimulation of resting human T cells results within minutes in an increase in ␤1-integrin-mediated adhesion to fibronectin. We have utilized the HL60 cell line to map critical residues within the CD2 cytoplasmic domain involved in CD2 regulation of integrin function. A panel of CD2 cytoplasmic domain mutants was constructed and analyzed for their ability to upregulate … Show more

“…Target cells (RMA, RMAS, RMA-Rae-1␦, RMAS-Rae-1␦, A20, or P815) were labeled with 51 Cr (100 Ci) for 1 h at 37°C and then washed. Labeled target cells (1 ϫ 10 4 ) were added to 96-well U-bottom plates containing activated CD8 or NK cells at the indicated ratios in a final volume of 200 l. After a 5-h incubation, the supernatants were collected and counted.…”

“…After electroporation, cells were incubated in complete medium for 16 -20 h at 37°C. The generation of stable P116 transfectants expressing ZAP-70 constructs was performed as previously described (18). Screening for transfectants was performed 20 -24 days following the electroporation by flow cytometry.…”

“…In T cells, however, CD2 clusters centrally or in an intermediate ring between the cSMAC and pSMAC (24). In T cells, CD2 can activate WASp (14), but may also activate ␤-integrins (25). In NK cells, however, CD2 Each cluster of bars represents the mean Ϯ SD of three to six independent experiments, and the entire figure summarizes Ϸ4,000 individual conjugates.…”
