Identification of a novel lncRNA induced by the nephrotoxin ochratoxin A and expressed in human renal tumor tissue

Abstract: Long non-coding RNAs represent a fraction of the transcriptome that is being increasingly recognized. For most of them no function has been allocated so far. Here, we describe the nature and function of a novel non-protein-coding transcript, named WISP1-AS1, discovered in human renal proximal tubule cells exposed to the carcinogenic nephrotoxin ochratoxin A. WISP1-AS1 overlaps parts of the fourth intron and fifth exon of the Wnt1-inducible signaling pathway protein 1 (WISP1) gene. The transcript is 2922 nucleo… Show more

“…Glucose consumption and lactate production were assessed using commercially available kits (Glucose [HK] assay kit, Sigma, Darmstadt, Germany and Lactate Reagent, Trinity Biotech, via Menarini Diagnostics, Berlin, Germany) according to the manufacturer's instructions [34]. Albumin was determined by ELISA (Bethyl Laboratories, Montgomery, TX, USA).…”

“…The latter play a role in gene expression regulation (as reviewed by Fernandes et al, 2019; Li et al, 2019 [50,51]) and are susceptible to environmental factors [52,53]. For instance, the transcript WISP1-AS1 was previously described as OTA-specifically induced and as involved in OTA-induced gene regulation and apoptosis [12]. In addition to providing a robust way of selecting relevant biological candidates, a network such as the one built in the present study also has the advantage of considering non-coding transcripts.…”

“…Despite this evidence, many published studies were based on experiments with higher OTA concentrations (micromolar range and even higher) and may, therefore, report unspecific toxicological reactions. We previously observed that nanomolar OTA triggers apoptosis [10], leads to gene expression dysregulation [11,12], influences signaling pathways [11,13], and can induce characteristic traits of interstitial nephritis in vitro [14]. However, the understanding of OTA nephrotoxicity following exposure to nanomolar concentrations is still limited.…”

“…Long noncoding RNAs (lncRNAs) belong to a class of regulatory noncoding RNAs with transcript lengths greater than 200 nucleotides and serve vital roles in transcriptional and posttranscriptional gene expression regulation [3,4]. Recently, dysregulation of lncRNAs has been found to be important in various human diseases, such as cancer, diabetes and cardiovascular diseases [5][6][7].…”
