Identification of a novel hybrid glycophorin gene encoding GP.Hop

Abstract: The GP.Hop phenotype is produced by a hybrid GP(B-A-B) protein caused by a DNA insertion of GYPA into GYPB. The composition of the hybrid protein is GPB(1-26)-GPpsiB(27-50)-GPA(51-58)-GPB(S)(59-103).

“…This GP hybrid is similar to GP.Mur and GP.Bun. In GP.HF, a 98 bp insert from exon 3 of GYP*A creates a GYP*(B-A-B) hybrid, which encodes a peptide differing from GP.Mur by five amino acid residues and from GP.Bun by six amino acid residues [32,33].…”

“…In addition to testing with antibodies to low-and high-prevalence MNS antigens, insight can be gained by the use of relatively simple serological tools such as tests with enzyme-treated RBCs. On intact RBCs, GPA is susceptible to cleavage by trypsin at amino acid residues 50 (31) and 58 (39) but is resistant to a-chymotrypsin cleavage whereas GPB is resistant to trypsin cleavage but sensitive to a-chymotrypsin at amino acid residue 51 (32) [26,41]. In the only known ENEV) proband, anti-ENEV caused a delayed transfusion reaction on several occasions [40].…”

“…The region around exon 3 (from intron 2 to 3) was reported to include a crossing-over hot spot responsible for some variant glycophorins (Vignal et al 1989;Kudo et al 1990;Huang et al 2000;Storry et al 2000). In a previous study (Sasaki et al 2000), the M102 and N102 alleles were considered to have been generated via recombination between the 5¢-region of M and the 3¢-region of N alleles around the hot spot.…”

“…Only two GP.Hop propositi are reported [289,319] . Like GP.Bun, GP.Hop red cells are also Mur + , MUT + , Hop + , and MINY + , but are Hil − and TSEN + (Table 3.10 ).…”
