Identification of a novel human doublecortin-domain-containing gene (DCDC1) expressed mainly in testis

Abstract: Mutations in the X-linked gene doublecortex (DCX) result in lissencephaly in males or subcortical laminar heterotopia (double cortex) in females. Recently, an evolutionarily conserved doublecortin (DC) domain important for microtubule binding and microtubule polymerization was defined according to detailed sequence analysis of DCX and DCX-like proteins. Subsequently we cloned a novel human cDNA that contained a DC domain during large-scale DNA sequencing of the human fetal brain cDNA library, and termed it dou… Show more

“…A similar strategy was used to analyze the metabolic effects of sequence variations in ANGPTL5 ( Figure 3A), a gene of unknown function that is not expressed in mice (13). A significant excess of NS variants was found in the lowest quartile of TG levels (n = 9) compared with the highest quartile (n = 1) (P = 0.022).…”

“…To this end, total human cord blood cells ( Figure 1A) were cultured in STIF medium containing Angptl5. 18 We seeded 2.5 ϫ 10 7 total cord blood cells at a density of 10 6 cells/mL medium. After 23 days of culture, the number of total cells in the presence of Angptl5 increased to 2.2 (Ϯ 0.3) ϫ 10 8 cells ( Figure 1A).…”

“…Thus, it is necessary to both understand the underlying molecular basis and develop effective preventive and therapeutic approaches. We and others have independently identified 7 angiopoietin-like proteins (Angptls), namely, Angptl1/angiopoietin-related protein 1 (ARP1)/angioarrestin, 1,2 Angptl2/ARP2, 3 Angptl3, 4,5 Angptl4/ARP4/PPARγ angiopoietin-related (PGAR)/fasting-induced adipose factor (FIAF)/hepatic fibrinogen/angiopoietin-related protein (HFARP), [6][7][8][9] Angptl5, 10,11 Angptl6/angiopoietin-like growth factor (AGF), 12 and Angptl7. 13 These Angptls are structurally similar to angiopoietins, which are characterized by a coiled-coil domain at the N-terminus and a fibrinogen-like domain at the C-terminus.…”
