Identification of a New, Unorthodox Member of the MAGE Gene Family

Abstract: Several tumor-associated antigen families, such as MAGE, GAGE/PAGE, PRAME, BAGE, and LAGE/NY-ESO-1, exist. These antigens are of particular interest in tumor immunology, because their expression, with exception of testis and fetal tissues, seems to be restricted to tumor cells only. We have identified a novel member of the MAGE gene family, MAGED1. Northern hybridization and RT-PCR demonstrated that the expression level of MAGED1 in different normal adult tissues is comparable to that in testis and fetal liver… Show more

“…Although the expression of the other CTSP-1 family members was also restricted among normal tissues, they were not frequently expressed in tumor cell lines and tumor samples. This fact has also been observed for other CT-antigen members of the MAGE and SSX families (17)(18)(19). A positive expression in normal testis was detected for CTSP-2 and -4, the first also being expressed in brain, fetal brain, and spinal cord among the 21 normal tissues analyzed (data not shown).…”

“…14 A cancer-testis antigen MAGE-E1 was identified using SAGE (serial analysis of gene expression) technique. 33 MAGE-D1, 34 SAGE and HAGE 35 were identified using RDA (representational difference analysis) method. Our study is the first report for identification of a novel melanoma antigen with melanocyte preferential expression using DNA chip and serum immunodetection.…”

“…3A. A putative human counterpart of mouse Dlxin-1 has been reported as MAGE-D1 (18). In the data base, SNERG-1 (GenBank TM accession number AF274043) from rat testis showed a high homology (96%) to mouse Dlxin-1, suggesting that SNERG-1 is a rat orthologue of mouse Dlxin-1 (Fig.…”
