Vibrio alginolyticus, Vibrio chlorea, and Enterococcus cloacae were the main bacterial pathogens investigated during recurrent episodes of mortalities among farmed flathead grey mullet (Mugil cephalus) in the Bahar El-Baqar area, Sharqia governorate, Egypt, during the summer season in 2021. Nevertheless, the contamination of water with heavy metals was evaluated. In this study, a total number of sixty moribund and freshly dead Mugil cephalus were examined clinically and upon post-mortem. Then, the bacterial isolates were subjected to bacteriological phenotypic examination, 16S rRNA gene sequencing, and antibiotic sensitivity testing. Histopathological screening and the immunity response of naturally infected M. cephalus were also implemented. In areas where bacterial strains were recovered in a high percentage, water contamination with Cu, Fe, and Ni was higher than the permissible limit. The results showed a high similarity (99–100%) to Vibrio and E. cloacae strains using phylogenetic analysis of 16S rRNA. Most isolates exhibited sensitivity to florfenicol, ciprofloxacin, and norfloxacin, while they were resistant to erythromycin and amoxicillin. Hemorrhage, oedema, hemosiderosis, and melanomacrophage activation in most affected tissues of infected M. cephalus were detected by light microscopy. Nevertheless, higher levels of AST and ALT, as well as serum inflammatory markers, CRP, and lysozyme activity, were shown in infected fish. There were higher MDA levels, along with a reduction of GSH, SOD, and catalase, in the gills and brain of infected M. cephalus, compared to the control fish. Our obtained data confirmed the high prevalence of Vibrio serovars and E. cloacae in infected grey mullets, describing their histopathological alterations, higher inflammatory response, and oxidative stress impact. These results could be useful indicators for understanding the pathogenesis of these pathogens in correlation with water parameter deterioration. Thus, the emergence of Vibrio and E. cloacae species in this study threatens the brackish water aquaculture industry in Egypt and poses a public health concern.

The supply of coconut crabs (Birgus latro Linnaeus, 1767) has been sourced from capture in the wild. Development efforts in aquaculture have succeeded in producing eggs. However, further development with larval maintenance failed because it was constrained by a low survival rate. This study aimed to identify the potential probiotic bacteria in coconut crabs. Coconut crab samples were collected from Moor Island, South Patani District, Central Halmahera Regency, North Maluku Province, Indonesia. The intestines of coconut crabs were taken as much as 1 g, then ground on a porcelain dish and added to Tri-Salt. The scouring liquid was taken in 0.1 ml and spread on a petri dish containing Nutrient Agar. The isolated used were marked with BL1, BL2, BL3, BL4, and BL5. Molecular identification was performed with sequencing and then analyzed using the Basic Local Alignment Search Tool (BLAST). The selection of isolates from the gastrointestinal tract of coconut crabs showed that the bacteria obtained were Enterobacter tabaci (BL1), Enterobacter hormaechei (BL2), Bacillus horneckiae (BL3), Pseudomonas stutzeri (BL4), and Acinetobacter variabilis (BL5). Further testing revealed that the bacteria A. variabilis (BL5) is a probiotic isolate in coconut crabs. Highlight Research The intestinal bacteria from coconut crabs identified were E. tabaci, E. hormaechei, B. horneckiae, P. stutzeri, and A. variabilis. The E. tabaci strain BL1, E. hormaechei strain BL2, B. Horneckiae strain BL3, and P. stutzeri strain BL4 are pathogenic bacteria. The A. variabilis strain BL5 is potential probiotics bacteria. The A. variabilis strain BL5 showed a better survival rate than other isolates.

Red swamp crayfish (Procambarus clarkii) have become one of the favorite aquatic products in China. The modern farming mode which uses antibiotics to prevent diseases could impact the bacteria in crayfish intestines. Here, we determined the distribution and antimicrobial resistance phenotypes of the selected culturable bacteria in crayfish intestines and characterized an isolate with last-line antibiotic resistance determinant. Totally, 257 strains were isolated from 115 crayfish. These strains were highly diverse, with Citrobacter sp. (n = 94, 36.6%) and Aeromonas sp. (n = 88, 34.2%) being dominant. Other isolates belonged to genera Pseudomonas, Myroides, Morganella, Klebsiella, Acinetobacter, Proteus, Enterobacter, Kluyvera, and Escherichia. Most strains from crayfish were susceptible to all tested antibiotics. None of the isolates carried last-line antibiotic resistance genes except one Escherichia coli isolate with blaNDM-5 was detected, which is the first report of blaNDM-5-positive E. coli isolate from red swamp crayfish. Whole-genome sequencing suggested it belonged to ST48 and carried several resistance genes. blaNDM-5 was located within an Tn3000-like transposon linked to an external 5 bp sequence (ACTAT) on both sides on a IncHI1B/HI1A/FIA multi-replicon plasmid. This transposon was considered to be acquired by replicative transposition mediated by IS3000. The emergence of bacteria with last-line antibiotic resistance genes in crayfish poses serious threat to public health since crayfish could act as a reservoir for the transfer of resistance to humans.
