Identification of a microRNA signature in endothelial cells with mechanical stretch stimulation

Abstract: The current study aimed to verify an miRNA signature in endothelial cells undergoing mechanical stretch stimulation. In the present study, microarray profiling was conducted in order to identify the differential expression of miRNAs in endothelial cells undergoing mechanical stimulation, compared with unstimulated endothelial cells. The microarray data was then validated by reverse transcription‑quantitative polymerase chain reaction. Genes and signaling pathways regulated by the miRNAs were investigated in si… Show more

“…Altered expression of miRNAs has been reported to occur on exposure to hypoxia, 23 mechanical stimulation, 24 and inflammation, 25 among other factors. The altered miRNA expression profile in turn may affect these processes.…”

“…The statistical power of this experimental design was estimated using the SSPA R package ( Iterson et al, 2013 ), which reached 0.63 and 0.76 for the HP vs LP and HD vs. LD, respectively ( Supplementary Figure S1 ). MiRNAs with a p -value <0.05 and log 2 (fold change)| >0.8 were assigned as DE ( Zheng et al, 2015 ). The expression patterns of DE miRNAs across four groups were performed using the k-mean method ( Ahmad and Dey, 2007 ).…”

“…miRNAs have been found to be critical modulators of endothelial homeostasis and link to vascular diseases ( Fernandez-Hernando and Suarez, 2018 ). miRNAs in ECs are mechanosensitive that response to mechanical stretch stimulation and exhibited obvious differential expression ( Kumar et al, 2014 ; Zheng et al, 2015 ). After 1h of mechanical ventilation with a tidal volume of 40ml/kg, the expression of 50 miRNAs increased more than 2-fold, and the expression of 15 miRNAs decreased by more than half ( Vaporidi et al, 2012 ).…”
