Identification of a Highly Conserved Domain on Phytochrome from Angiosperms to Algae

Abstract: A monoclonal antibody (Pea-25) directed to phytochrome from etiolated peas (Pisum sativam L., cv Alaska) binds to an antigenic domain that has been highly conserved throughout evolution. Antigenic crossreactivity was evaluated by immunoblotting sodium dodecyl sulfate sample buffer extracts prepared from lyophilized tissue samples or freshly harvested algae. Pea-25 immunostained an approximately 120-kilodalton polypeptide from a variety of etiolated and green plant tissues, including both monocotyledons and dic… Show more

“…Western blot analysis using the monoclonal antibody (P25) generated against pea phyA (Cordonnier et al, 1986) was performed as described by Heyer and Gatz (1992b). For the western blot analysis shown in Figure 4B a different set of antibodies was used.…”

“…In algae, phytochrome detected in some fresh-water and marine chlorophyta (Taylor & Bonner 1967;Cordonnier et al 1986), in red algae (López-Figueroa & Niell 1988;López-Figueroa et al 1989), in brown algae (López-Figueroa et al 1989), in cyanobacteria (Scheibe 1972;Björn & Björn 1976, 1980, and in phyco-and photobionts of lichens (Czeczuga et al 2008). Phytochrome also was detected in Treobuxia sp.…”

“…Cross-reacting proteins had been detected with monoclonal antibodies directed against pea phytochrome in the green algae Mougeotia, Mesotaenium and Chlamydomonas (Cordonnier et al, 1986). More recently, phytochrome-like proteins were detected in Dunaliella and Chlorella (Ruyters et al, 1991) and in several Rhodophyceae, Phaeophyceae and Chlorophyceae using monoclonal antibodies raised against phytochrome from etiolated Avena saliva and Zea mays (L6pez-Figueroa et al, 1989a).…”
