Identification of a Distinct Small Cell Population from Human Bone Marrow Reveals Its Multipotency In Vivo and In Vitro

Abstract: Small stem cells, such as spore-like cells, blastomere-like stem cells (BLSCs), and very-small embryonic-like stem cells (VSELs) have been described in recent studies, although their multipotency in human tissues has not yet been confirmed. Here, we report the discovery of adult multipotent stem cells derived from human bone marrow, which we call StemBios (SB) cells. These isolated SB cells are smaller than 6 ìm and are DAPI+ and Lgr5+ (Leucine-Rich Repeat Containing G Protein-Coupled Receptor 5). Because Lgr5… Show more

“…The bone marrow is the most widely studied stem cell niche in the adult, however many other tissues and fluids such as the dental pulp, cord blood, breastmilk, the basement membrane of the seminiferous tubules, and the endometrium contain stem cells with pluripotent features [ 19 , 20 , 21 , 22 , 23 ]. Mesenchymal/stromal stem cells (MSCs) from the bone marrow are defined by their ability to differentiate into osteoblasts, adipocytes and chondrocytes and express specific markers including CD44, CD63, CD105 and CD146 [ 24 ].…”

“…Despite having smaller sizes than OSCs, VSELs express nuclear OCT4 rather than cytoplasmic OCT4 in OSCs, which defines their pluripotent nature [101]. These pluripotent cells can be isolated from a number of other adult cell sources such as bone marrow [102], bone [103], umbilical cord blood [104], and peripheral blood [105], and they can be used to derive OLCs when induced under suitable culturing conditions. Due to the quiescent nature of VSELs, they can avoid nuclear and mitochondrial mutations and can therefore be used as safer therapeutic agents in reproductive medicine [106].…”

“…SB cells ® were collected from patients using SB cells ® purification protocol as mentioned in our previous publication. SB cells ® were then injected back to patients following the guidelines described in our procedural IRB (IRB protocol number SB-IN-4112) [1].…”

“…Previous publication of flow cytometry analysis shows the presence of SB cells ® in the small cell like region (3-7μm) devoid of white blood cells and red blood cells. SB cells ® are less than 6μm in diameter, expresses Lgr5+ and CD349+ but not CD133-, CD45-, and CD66e-; thereby, concluding properties dissimilar to very-small embryonic-like stem cells (VSELs), hematopoietic stem cells (HSCs), and blastomerelike stem cells (BLSCs) [1,2].…”
