Identification of a cellular ligand for the natural cytotoxicity receptor NKp44

Abstract: Key Points• The cellular ligand of the NKp44L is a novel isoform of the mixed-lineage leukemia-5 protein.• NKp44L is not expressed on healthy cells, but on tumor and transformed cells, rendering them more sensitive for the NK cytotoxicity. With an array of activating and inhibitory receptors, natural killer (NK) cells are involved in the eradication of infected, transformed, and tumor cells. NKp44 is a member of the natural cytotoxicity receptor family, which is exclusively expressed on activated NK cells. Here… Show more

“…3G). Differently from K562 target cells, HEK 293T cell line expresses ligands for both NKp44 and NKG2D (38,49,50). Therefore, the degree of CD107a pos M1-primed NK cells was significantly reduced in the presence of NKp44 masking and was restored to almost basal levels when NKp44 and NKG2D were blocked together (Fig.…”

“…Although NCRs were found to bind some viral glycoproteins, their ligands on tumour cells have only partially been defined 10,11 . B7-H6 and, very recently, a novel isoform of the mixed-lineage leukemia (MLL5) protein have been shown to be expressed on a large panel of tumour cells 12,13 and are recognized by NKp30 and NKp44 receptors, respectively. Although a role of NK cells in anti-tumour immunity has been established, such a role for other ILC populations remains largely unexplored.…”
