Identification and Validation of Reference Genes for Seashore Paspalum Response to Abiotic Stresses

Abstract: Seashore paspalum (Paspalum vaginatum) is among the most salt- and cadmium-tolerant warm-season perennial grass species widely used as turf or forage. The objective of this study was to select stable reference genes for quantitative real-time polymerase chain reaction (qRT-PCR) analysis of seashore paspalum in response to four abiotic stresses. The stability of 12 potential reference genes was evaluated by four programs (geNorm, NormFinder, BestKeeper, and RefFinder). U2AF combined with glyceraldehyde-3-phosph… Show more

“…In this study, U2AF displayed the highest stability in Cd, Hg, Cr, and As treated leaves as well as in Pb, Hg, Cr, and As treated roots of switchgrass. U2AF has been reported in other studies to be the most stable reference gene, such as in Pinus massoniana L., at different stages post-inoculation by nematode [35] and roots/leaves of Paspalum vaginatum Sw. under Cd and cold stress [36]. In the roots of switchgrass subjected to Cd treatment, CYP5 and UBQ1 were determined to be the most appropriate internal reference genes.…”

“…Not surprisingly, each algorithm gave a different ranking. According to previous research, the use of multiple reference genes is preferable to gain a more accurate and reliable result ( Vandesompele et al, 2002 ; Martins et al, 2016 ; Liu et al, 2017 ). The optimal number of reference genes to achieve normalization can be calculated using geNorm, where the lowest number of gene combinations which causes the threshold value to fall below 0.15 indicates the number of reference genes which will be sufficient for normalization ( Vandesompele et al, 2002 ).…”

“…The RefFinder software comprehensively ranks the candidate reference genes based on the geometric mean (GM) values of the results from the four different statistical algorithms described above ( Xie et al, 2012 ). With the help of these five statistical algorithms, much research on the validation of reference genes under different experimental conditions has been reported ( Liu et al, 2017 ; Sadangi et al, 2017 ; Sprang et al, 2017 ; Mughal et al, 2018 ).…”

“…ACT and GAPDH exhibited variable expression in leaves of bermudagrass ( Cynodon dactylon ) under NaCl and PEG stress [ 18 ]. Likewise, both genes performed poorly in seashore paspalum ( Paspalum vaginatum ) exposed to drought and salinity [ 19 ]. Furthermore, GAPDH was reported to be least stable in annual ryegrass ( Lolium multiflorum ) under abovementioned stresses [ 16 ], while ACT was least stable in durum wheat ( Triticum durum ) subjected to drought [ 33 ].…”
