There is evidence supporting the notion that Candida albicans (C. albicans) indeed contributes to human cancers. Interestingly, the efficacy of C. albicans in improving Colorectal cancer (CRC) has been confirmed. This study primarily explores the paradox of whether C. albicans promotes or inhibits the development of CRC, focusing on its metabolites mixture for relevant arguments. This study identified a total of 214 differentially expressed genes. A prognostic model containing 5 specific mRNA markers, namely EHD4, LIME1, GADD45B, TIMP1, and FDFT1, was constructed. C. albicans metabolites mixture reduced CRC cell activity. qRT-PCR results showed that compared to normal colonic epithelial cells, LIME and EHD4 were downregulated in CRC cells, while FDFT1 expression was significantly upregulated. Notably, the TIMP1 gene was significantly upregulated in HT29 cells, while it was significantly downregulated in HCT116 cells. Furthermore, post-intervention analysis showed a significant decrease in gene expression levels in HT29 cells, while the expression of TIMP1, EHD4, and GADD45B increased in HCT116 cells, with LIME and other CRC cells showing a corresponding decrease in expression. In NCM460 normal colonic epithelial cells, the expression levels of GADD45B, TIMP1, and FDFT1 genes were significantly upregulated, while the expression levels of LIME and EHD4 showed a significant downward trend. After metabolite intervention, the invasion and migration capabilities of NCM460 cells, HT29 cells, and HCT116 cells decreased. Additionally, quantitative measurement of eATP levels after intervention showed a significant increase (P < 0.01). This study's prognostic model opens up a new paradigm for prognostic assessment in CRC. The metabolites mixture of C. albicans play a protective role in the onset and progression of CRC, exhibiting dynamic interactions with cellular energetics.

To explore the mechanism of necrotic effect of nourishing cells in the context of genital tract infection in premature rupture of membranes (PROM). One hundred eight patients with PROM treated at our hospital from June 2020 to June 2022 were selected as the PROM group. Simultaneously, 108 cases of normal full-term pregnant women were chosen as the control group. Western blot analysis was performed to measure the relative expression levels of cysteinyl aspartate specific proteinase-1 (Caspase-1), cysteinyl aspartate specific proteinase-3 (Caspase-3), nucleotide-binding oligomerization domain-like receptor family pyrin domain containing 3 (NLRP3), and interleukin (IL)-1β proteins, which are associated with necrosis of placental nourishing cells, in the placenta of both groups. TUNEL staining was used to detect the number of apoptotic placental nourishing cells. The differences in necrotic factors of placental nourishing cells were analyzed between full-term and preterm cases in the PROM group, as well as among patients with different genital tract infections. The apoptotic count of placental nourishing cells in the PROM group was 58.46 ± 11.26 cells/field, which was markedly higher than that of the control group (P < .05). The relative expression levels of the necrotic factors Caspase-1, Caspase-3, NLRP3, and IL-1β proteins in placental nourishing cells of the PROM group were 1.32 ± 0.26, 1.19 ± 0.30, 1.29 ± 0.28, and 1.23 ± 0.24, respectively. These values were significantly higher than those of the control group (P < .05). The relative expression levels of the necrotic factors Caspase-1, Caspase-3, NLRP3, and IL-1β proteins in placental nourishing cells were compared between full-term and preterm patients in the PROM group (P > .05). The relative expression levels of the necrotic factors Caspase-1, Caspase-3, NLRP3, and IL-1β proteins in placental nourishing cells were higher in patients with multiple genital tract infections compared to those with single infections or no infections in the PROM group (P < .05). PROM is associated with a significant upregulation of placental nourishing cell apoptosis and necrotic factors, including Caspase-1, Caspase-3, NLRP3, and IL-1β proteins. This upregulation is correlated with the presence of genital tract infections.

Ubiquitination-related genes (URGs) exerted a crucial part in a variety of human disease disorders; however, their association with pancreatic adenocarcinoma (PAAD) had yet to be clearly described. We aimed to comprehensively characterize the contributions of URGs in PAAD through in silico analysis and experimental validation, and then identified a robust mRNA-lncRNA-based molecular prognostic panel for patients with PAAD using bulk RNA-sequencing and single-cell RNA-sequencing data. Initially, we collected the multi-omics data from TCGA platform to depict a comprehensive landscape of URGs in pan-cancer. Furthermore, we were accurate to PAAD for in-depth analysis. Significant differences of the activation of ubiquitination pathways and the expression of URGs were detected between normal and malignant cells. Unsupervised hierarchical clustering determined two PAAD subtypes with distinct clinical outcomes, ubiquitination pathway activities, immune microenvironment, and functional annotation characteristics. The expression profiles of ubiquitination-associated mRNAs and lncRNAs in the training and validation datasets were utilized to develop and verify a novel ubiquitination-related mRNA-lncRNA prognostic panel, which had a satisfied prediction efficiency. Our ubiquitination-associated model could function as an effective prognostic index and outperformed four other recognized panels in evaluating PAAD patients’ survival status. Tumor immune microenvironment, mutation burden, and chemotherapy response were intensively explored to demonstrate the underlying mechanism of prognostic difference according to our panel. Our findings also revealed that FTI-277, a farnesyltransferase inhibitor, had a better curative effect in high-risk patients, while MK-2206, an Akt allosteric inhibitor, had a superior therapeutic effect in low-risk patients. The real-time PCR results uncovered the RNA expression of AC005062.1 in all the three PAAD cell lines was elevated several thousandfold. In conclusion, our URGs-based classification panel could be triumphantly served as a prediction tool for survival evaluation in patients with PAAD, and the genes in this panel could be developed as a potential target in PAAD therapy.
