Identification and transcript analysis of two glutamate decarboxylase genes, CsGAD1 and CsGAD2, reveal the strong relationship between CsGAD1 and citrate utilization in citrus fruit

Abstract: Glutamate decarboxylase (GAD, EC 4.1.1.15) has been suggested to be a key, regulatory point in the biosynthesis of γ-aminobutyrate (GABA) and in the utilization of citric acid through GABA shunt pathway. In this study we discovered two GAD genes, named as CsGAD1 and CsGAD2, in citrus genome database and then successfully cloned. Both CsGAD1 and CsGAD2 have a putative pyridoxal 5-phosphate binding domain in the middle region and a putative calmodulin-binding domain at the carboxyl terminus. Gene structure analy… Show more

“…qRT-PCR was performed in a 10 μL reaction volume that contained 5 μL Thunderbird TM SYBR qPCR Mix (TOYOBO, JAPAN), 1 μL of cDNA, 1 μM gene-specific primers and 3 μL ddH 2 O. Reactions started with an initial incubation at 50°C for 2 min and 95°C for 10 min and were then subjected to 40 cycles of 95°C for 15 s and 60°C for 60 s (Liu et al, 2014). The Livak (Livak and Schmittgen, 2001) method was employed to calculate gene relative expression levels.…”

“…Each pair of fruits was grown on the identical fruit-bearing shoot, and the fruits were similar in size. The ABA injection was performed according to Liu et al (2014). In each pair of fruits, one fruit was injected with an ABA solution, and one fruit, the control, was injected only with the solvent (without the ABA).…”

“…SHM7 , also known as AT1G36370, encodes the specific serine hydroxy methyltransferase 7, which further contributes to catalyzing the interconversion of serine and glycine [71]. Recent publications confirm that the main function of SHM7, pyridoxal phosphate binding, may contribute to the development and maturation of fruit in citrus, implying its potential role in various plant subtypes [72, 73]. Furthermore, a specific mutant screening of Arabidopsis thaliana , based on high-throughput HPLC-MS/MS assay has confirmed that, together with another enzyme threonine aldolase, the serine hydroxy methyltransferase may play a specific role during the development and maturation of fruit, affecting seed nutritional quality [74].…”
