Identification and quantitation of indole-3-acetic and abscisic acids in the cambial region of Quercus robur by combined gas chromatography-mass spectrometry

Abstract: A technique is described for the rapid purification of indole-3-acetic acid (IAA) and abscisic acid (ABA) from small quantities of the basal trunk cambial region tissues of 34-year-old trees of Quercus robur L. Mass spectra of IAA and ABA obtained from the purified extracts provide the first reported unequivocal identification of endogenous IAA and ABA in the cambial region of a hardwood species. By use of the combined gas chromatography-mass spectrometry technique of multiple ion monitoring, with deuterated a… Show more

“…Frozen samples were lyophilized and stored at )70°C until extraction. The level of endogenous ABA was quantified following the method of Browning and Wignall (1987), Qi et al (1998), and Kamboj et al (1999). Plant samples were extracted with 30 ml of extraction solution containing 95 % isopropanol, 5 % glacial acetic acid, and 100 ng of [(±)-3,5,5,7,7,7-d 6 ]-ABA.…”

“…The ABA content was analyzed using the method of Browning and Wignall (1987), Qi et al (1998), and Kamboj et al (1999) with modifications. The powdered seeds were extracted with 30 of solution containing 95% isopropanol and 5% glacial acetic acid, and 50 ng of [(±)-3,5,5,7,7,7-d6]-ABA standard was added into extracts.…”
