BackgroundOphiostoma novo-ulmi is the causative agent of Dutch elm disease (DED). It is an ascomycetous filamentous fungus that ranks as the third most devastating fungal pathogen in Canada. The disease front has spread eastward and westward from the epicentre in Ontario and Quebec and is threatening elm populations across the country. Numerous mitigation strategies have been tried to eradicate this pathogen, but success has thus far been limited. An alternative approach might utilize double-stranded RNA (dsRNA) mycoviruses which have been reported to induce hypovirulence in other fungi.MethodsUsing a modified single primer amplification technique (SPAT) in combination with chromosomal walking, we have determined the genome sequence of two RdRp encoding dsRNA viruses from an O. novo-ulmi isolate (93–1224) collected from the disease front in Winnipeg.ResultsWe propose that these viruses, which we have named OnuMV1c and OnuMV7 based on sequence similarity to other Ophiostoma mitoviruses, are two new members of the genus Mitovirus in the family Narnaviridae.ConclusionsThe discovery of such dsRNA elements raises the potential for engineering these viruses to include other genetic elements, such as anti-sense or interfering RNAs, to create novel and highly specific biological controls. Naïve fungal hosts could be infected with both the engineered molecule and a helper mitovirus encoding an RdRp which would provide replication capacity for both molecules.

Abstract. Various Resumen. Se utilizan varios métodos para realizar la extracción y purificación de ARN de doble cadena (dsRNA) largo de plantas y hongos. Los protocolos más utilizados consisten en la extracción de fenol y la unión selectiva de dsRNA a la celulosa. Los dsRNAs virales purificados de plantas y hongos se han analizado por electroforesis con gel y se han utilizado como herramienta complementaria para identificar virus. Asimismo, los dsRNAs se han utilizado como reactivos en PCR de transcriptasa reversa, clonación molecular, preparación de sondas y secuenciación de virus ARN. Se han descubierto muchos virus ARN y moléculas subvirales que infectan plantas y hongos utilizando protocolos de extracción de dsRNA. Las recientes mejoras a los métodos de extracción de dsRNA han aumentado la eficiencia y la rentabilidad. Se ha comprobado que el dsRNA viral se puede utilizar como reactivo inicial para la secuenciación de próxima generación de genomas virales.

Ever since their discovery just about 56 years ago in the cultivated mushroom , many more viruses infecting fungi have been identified in a wide range of fungal taxa. With mostly being asymptomatic, especially the ones that are detrimental to their phytopathogenic hosts are intensively studied due to their considerable importance in developing novel plant protection measures. Contrary to the rapid accumulation of notable data on viruses of plant pathogenic microfungi, much less information have hitherto been obtained in regards to the viruses whose hosts are macrofungi. According to the current literature, only more than 80 distinct viruses bearing either linear dsRNA or linear positive sense ssRNA genome and infecting a total number of 34 macrofungal species represented with four and 30 have been identified so far. Among these 34 macrofungal species, 14 are cultivated edible and wild edible mushroom species. According to the 10th ICTV (International Committee on Taxonomy of Viruses) Report, macrofungal viruses with linear dsRNA genome are classified into five families (, ,, and) and macrofungal viruses with linear positive sense ssRNA genome are classified into seven families (, ,, ,, and). In this review, following a brief overview of some general characteristics of fungal viruses, an up to date knowledge on viruses infecting macrofungal hosts were presented by summarizing the previous, recent and prospective studies of the field.
