Identification and Genomic Characterization of a New Virus ( Tymoviridae Family) Associated with Citrus Sudden Death Disease

Abstract: Citrus sudden death (CSD) is a new disease that has killed approximately 1 million orange trees in Brazil. Here we report the identification of a new virus associated with the disease. RNAs isolated from CSD-affected and nonaffected trees were used to construct cDNA libraries. A set of viral sequences present exclusively in libraries of CSD-affected trees was used to obtain the complete genome sequence of the new virus. Phylogenetic analysis revealed that this virus is a new member of the genus Marafivirus. Ant… Show more

“…Various researches have suggested the pathogen based theory. For example, Maccheroni et al (2005) suggest that the pathogen could be a new strain of citrus tristeza or a new virus belonging to the family Tymoviridae (gender Marafivirus) and transmitted by insects.…”

“…Two viruses similar to plants infected with CSD, the virus CTV (Citrus tristeza virus, family Closteroviridae) and a new type virus belonging to the family Tymoviridae, as well as their association with the disease has been studied (Coletta Maccheroni et al, 2005;Terencio et al, 2005). Up to now, the diagnostics of CSD is based on signs found in sick plants.…”

“…Search for infectious agents in CSD-symptomatic trees including fungi, exogenous and endogenous bacteria and phytoplasmas, and viroids produced negative results (Bassanezi et al, 2003;Román et al, 2004). Only two virus, CTV and a new virus Tymoviridae, tentatively called Citrus sudden death associated virus (CSDaV), have been found in CSDaffected trees, and their association with the disease has been studied (Coletta Filho et al, 2005;Maccheroni et al, 2005). However, the extreme variability and complexity of CTV and the very low concentration of CSDaV make the CSD etiology very difficulty to be proven.…”

“…However, CTV and a new marafivirus (Tymoviridae) provisionally called Citrus sudden death associated virus (CSDaV) have been found in diseased trees. The association of CSD affected trees with CTV variants (Coletta-Filho et al, 2005;Maccheroni et al, 2005) and the new marafivirus (Jesus Junior et al, 2005;Maccheroni et al, 2005) has been studied, but the role these viruses play in CSD is still unclear and Koch's postulates have not yet been fulfilled.…”
