The 14-3-3 gene plays important role in many biological processes, including cell survival, apoptosis, and signal transduction. However, function of the 14-3-3 homologous gene in Musca domestica remains unclear. Here, we identified and characterized the 14-3-3ζ of M. domestica. We found that Md14-3-3ζ gene was highly homologous with other close insects. The qRT–PCR analysis revealed that the Md14-3-3ζ was highly expressed in adults, and was expressed predominantly in hemocytes and fat body. Meanwhile, the expression of Md14-3-3ζ was up-regulated after injecting Escherichia coli and Staphylococcus aureus. Moreover, the recombinant protein rMd14-3-3ζ strongly inhibits the growth of E. coli and S. aureus. Notably, the rMd14-3-3ζ inhibits E. coli and S. aureus by permeating the cell membrane. Taken together, our findings suggested that Md14-3-3ζ is involved in the immune response against bacteria through damaging the cell membrane.

Pest management in farm animals is an important action to contain economic damage to livestock production and prevent transmission of severe diseases to the stock. The use of chemical insecticides is still the most common approach followed by farmers; however, avoiding possible toxic effects on animals is a fundamental task for pest control measures compatible with animal well‐being. Moreover, legal constraints and insurgence of resistance by target species to the available insecticidal compounds are increasingly complicating farmers' operations. Alternatives to chemical pesticides have been explored with some promising results in the area of biological control or the use of natural products as sprays. The application of RNA interference techniques has enabled the production of new means of pest control in agriculture, and it is opening a promising avenue for controlling arthropod pests of livestock. Transcript depletion of specific target genes of the recipient organisms is based on the action of double‐strand RNAs (dsRNA) capable of impairing the production of fundamental proteins. Their mode of action, based on the specific recognition of short genomic sequences, is expected to be highly selective towards non‐target organisms potentially exposed; in addition, there are physical and chemical barriers to dsRNA uptake by mammalian cells that render these products practically innocuous for higher animals. Summarising existing literature on gene silencing for main taxa of arthropod pests of livestock (Acarina, Diptera, Blattoidea), this review explores the perspectives of practical applications of dsRNA‐based pesticides against the main pests of farm animals. Knowledge gaps are summarised to stimulate additional research in this area.

BACKGROUNDThe white‐backed planthopper (WPH), Sogatella furcifera (Horváth), is a destructive rice pest with strong reproductive capacity. To gain insights into the roles of chitinases in the reproductive process of this insect species, this study represents the first‐ever endeavor to conduct an in‐depth exploration into the reproductive functions of four chitinase genes.RESULTSIn this study, it was observed that four chitinase genes were expressed in female adults, with a relatively high expression level in the ovaries. SfCht2 and SfIDGF1 were highly expressed during later ovarian development. while SfENGase increased and then decreased with ovarian development. SfCht2, SfCht6‐2 and SfENGase were highly expressed in fat body on the first and second days after eclosion, whereas SfIDGF1 highest on day 7. Compared with control group, Silencing four chitinase genes inhibited ovarian development and significantly shortened the oviposition period of S. furcifera, reducing egg‐laying capacity but not affecting egg hatching. The detection demonstrated that the expression levels of SfVg, SfVgR and 70–90% juvenile hormone (JH) signaling pathway‐related reproductive genes was significantly down‐regulated. Moreover, SfCht6‐2 and SfENGase significantly affected the expression levels of Target of Rapamycin (TOR) signaling pathway genes. SfENGase had the ability to impact nutrient signaling pathways and fatty acid metabolism, repressing vitellogenin synthesis and ultimately influencing ovarian development of S. furcifera.CONCLUSIONSOverall, this study provides insight into the function of chitinases in insect fecundity and is of great significance for enriching the cognition of insect chitinase function. They will become the suitable target genes for controlling the most destructive rice planthoppers. © 2023 Society of Chemical Industry.
