Identification and expression analysis of major histocompatibility complex IIB gene in orange‐spotted grouper Epinephelus coioides

Abstract: In this study, complementary DNA (cDNA) and DNA sequences of major histocompatibility complex (MHC) class IIB genes (mhcIIB) were cloned from orange-spotted grouper Epinephelus coioides. The gene structure of E. coioides mhcIIB consists of five exons and four introns, and its deduced amino acid sequence length is 249 amino acids, including a signal peptide, a peptide-binding region, an IGC1 domain, a transmembrane region and a cytoplasmic tail. A phylogenetic study showed that E. coioides mhcIIB shared 32.0-79… Show more

“…However, the expression levels of MHC-II were not significantly different among each group before and after L. crocea was immunized with rCiSA32.6t in this work, which is in agreement with earlier reports on inactivate theront-immunized E. coioids (Dan et al 2013 ). It is speculated that MHC-II may not be involved in the presentation of the C. irritans surface antigen CiSA32.6t or MHC-II antigen presentation may occur at an early stage (1 to 7 days) after immunization with C. irritans vaccine (Deng et al 2020 ; Lu et al 2012 ). IgM is discovered to function as the front-line molecules of host defense against pathogen infection and plays an important role in humoral immunity by binding antigens and activating the complement system (Mashoof and Criscitiello 2016 ).…”

“…Primary spleen cells from T. nigroviridis were obtained as previously described, with minor modifications [50]. Briefly, tissue culture medium (TCM) was prepared from RPMI-1640 medium (Life Technologies, Gaithersburg, MD, USA) by adding 10% fetal bovine serum (Life Technologies), 2 mM L-glutamine (Sigma, St. Louis, MO, USA), and 1% penicillin/streptomycin (Sigma).…”
