Identification and Developmental Changes of Aromatase and Estrogen Receptor Expression in Prepubertal and Pubertal Human Adrenal Tissues

Abstract: Our results suggest that estrogens produced locally in adrenal medulla would play a role in zona reticularis functional differentiation through ERbeta. The cP450arom and GPR30 expression in subcapsular ZG, colocalizing with a high-cell proliferation index, previously reported, suggests a local GPR30-dependent estrogen action in proliferation and migration of progenitor adrenal cells.

“…While such low levels of expression could not significantly affect circulating levels of adrenally sourced corticosteroid, the abundance of MR and GR within hippocampus and cerebellum suggest a paracrine or autocrine mode of action for any steroid synthesised within the CNS. RT-PCR of adrenal cDNA also detected transcription of the aromatase (CYP19) gene, confirming the recent study by Baquedano et al (2007) who detected aromatase mRNA in normal adrenal medulla and zona glomerulosa and confirmed the enzyme's expression by immunohistochemistry.…”

“…This receptor is highly expressed in the ZG and medulla of the human adrenal gland [34], in cultured bovine adrenal medullary cells [35] and in adrenal medulla of GPR30-LacZ reporter mice [36]. In the male and female rats adrenal medulla and cells within the ZG of the adrenal cortex, high levels www.fhc.viamedica.pl of GPR30 mRNA and GPR30-like immunoreactivity were observed [37].…”

“…In contrast to animals, numerous data are available on aromatase expression in human adrenals. Aromatase expression has been observed in the human fetal and adult normal adrenal gland [29,34,46]. However, expression of aromatase gene is very high in adrenocortical tumors, especially those secreting estrogens [29,47,48].…”

“…Stojković R, et al Age and sex differences in genome damage between prepubertal and adult mice after exposure to ionising radiation Arh Hig Rada Toksikol 2016;67:297-303 Sex-and age-related differences in the effects of ionising radiation are affected by different levels of oestrogen receptors in tissues during prepuberty in comparison with adults, as reported in human adrenal tissue (25). Increased levels of oestrogen two months after exposure to ionising radiation were described in prepubertal mice (5).…”
