Identification and Development of Subtypes With Poor Prognosis in Pan-Gynecological Cancer Based on Gene Expression in the Glycolysis-Cholesterol Synthesis Axis

Abstract: Objective: Metabolic reprogramming is an important biomarker of cancer. Metabolic adaptation driven by oncogenes allows tumor cells to survive and grow in a complex tumor microenvironment. The heterogeneity of tumor metabolism is related to survival time, somatic cell-driven gene mutations, and tumor subtypes. Using the heterogeneity of different metabolic pathways for the classification of gynecological pan-cancer is of great significance for clinical decision-making and prognosis prediction. Methods: RNA sequ… Show more

“…Genes involved in glycolysis-cholesterol synthesis axis have been associated with immune infiltration and prognosis in ovarian, cervical, endometrial, breast, and pancreatic cancers, indicating that there is an interaction between the TME and tumor metabolism (24,44,45). Glucose deprivation attenuates the anti-tumor immune response triggered by Cytotoxic T Lymphocytes (CTLs) in glycolytic-dependent tumor cells, whereas checkpoint antagonists, such as anti-PD1 or anti PDL1 antibodies, provide glucose to CTLs by inhibiting glycolysis (46,47).…”

“…Survival ROC package was used to determine the best cutoff value of new lesions [17]. Time ROC package was used to present 1-year time-dependent receiver operating characteristic (ROC) curves [18]. All statistical analyses were conducted using SPSS 23.0 and R 4.1.0.…”

“…We performed GSEA to investigate signaling pathways regulated by the different molecular subtypes by using all candidate gene sets in the HALLMARK database ( Liberzon et al, 2015 ). The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis on the 2,443 genes were performed by the WebGestaltR package ( Liao et al, 2019 ; Wang et al, 2021a ). The p values smaller than 0.05 was determined as statistically significant.…”
