Identification and cloning of the porcine ROSA26 promoter and its role in transgenesis

Abstract: Several promoters from different origins have been used to drive transgene expression in pigs with variable results. In this study, we identified porcine ROSA26 promoter located on chromosome 13. Porcine ROSA26 promoter was cloned and its activity was examined using enhanced green fluorescence protein (EGFP) as a reporter. EGFP expression was detected in adult liver derived cells (LDC) by flow cytometry and confocal microscopy after transient transfection. Stable LDC expressing EGFP were established and used a… Show more

“…The sequence for synthesis of HDR and ROSA26 was first confirmed by Sanger sequencing. Previously published sequencing for the promoter region and exon 1 of the porcine ROSA26 locus was used to complete the gene template (54). The target sequence was interrogated against the porcine genome in the NCBI database using the Basic Local Alignment Search Tool (BLAST) (https://blast.ncbi.nlm.nih.gov/Blast.cgi) and a sequence similar to the NC_010455.4 (72802131 to 72802975) located at chromosome 13 in the Sus scrofa genome (S. scrofa).…”

“…The porcine ROSA26 locus has previously been targeted (Li S. et al, 2014), revealing ubiquitous expression of inserted transgenes in living pigs and that the strength of the porcine ROSA26 promoter is comparable to the H-2K b promoter (Li P. et al, 2014).…”
