Identification and Characterization of the Bovine Herpesvirus 5 US4 Gene and Gene Products

Abstract: The BHV-5 strain N569 (BHV-5/N569) homolog to the BHV-1 US4 gene was sequenced and characterized. RNA analyses showed that a 1.8-kb mRNA which contains the BHV-5/N569 US4 open reading frame initiates 55 nucleotides upstream from the predicted translational start codon and terminates 17 nucleotides downstream from the consensus sequence for polyadenylation. Comparison of the deduced amino acid sequences of the predicted US4 encoded proteins of BHV-5/N569 and BHV-1 strain Schönböken (BHV-1/Schö) revealed 75% ide… Show more

“…The U S region, extending from positions 116733 to 126280, contains eight genes (US1.67, 2 to 4, and 6 to 9), four of which have been previously sequenced (1,20,22,31). BHV-5 U S genes exhibit 69 to 98% amino acid identity (average, 79%) and overall less conservation to BHV-1 homologues than those within the U L region (84% average amino acid identity) (58).…”

“…The US2 homologue in HSV-1 and HSV-2 seems dispensable in tissue culture and is not involved in HSV-2 neuropathogenesis in mice (52,61). BHV-5 US4 encodes a glycosaminoglycan-associated protein (31) and is 72% identical to BHV-1 gG, which is involved in cell-cell virus transmission in vitro (69) and in prevention of apoptosis in certain cell lines (68). BHV-5 US8 encodes gE, which is important for neurovirulence in rabbits.…”

“…In addition, secreted gG can be found as protein species ranging from 90 to 240 kDa when linked to GAGs (27,33). BHV-1 gG is nonessential for viral growth but essential for cell-to-cell spread in bovine kidney cells (47,58).…”

“…1C). The upstream fragment that was chosen was the 1.8 kb NruI fragment that starts 149 nt upstream of the end of the gG coding region, covers the complete gD coding region and ends at the start of the gI coding region (5,8). The downstream fragment that was chosen was 1.6 kb NotI fragment that starts at the end of the US9 coding region, covers the partial US1.67 gene and ends in the terminal repeat region (5).…”
