Identification and characterization of the human type II collagen gene (COL2A1).

Abstract: The (7,8). To approach these questions, a variety of cDNA and genomic collagen clones from a number of species have been isolated, including the human al(I) (9, 10) and a2(I) genes (11, 12). We previously reported the isolation of the genomic clone CosHcoll from a human placental cosmid library (13), using the chicken al(I) cDNA clone pCg54 as a probe (14). This cosmid clone contains a 36-kilobase (kb) insert and crosshybridizes with collagen al(I) mRNA. However, the amino acid sequence derived from 1 kb of t… Show more

“…The Col2a1 gene encoding the major cartilage specific (Type II) collagen is expressed fairly early and very robustly within the chondrogenic lineage, as well as in certain other tissues, such as the notochord and the apical ectodermal ridge (AER) of the limb bud, albeit at much lower levels (Cheah et al, 1985(Cheah et al, , 1991Lui et al, 1995). Furthermore, the regulatory elements driving expression of the Col2a1 gene have been functionally dissected and very well characterized in transgenic mouse lines.…”

“…In each primer set, the nucleotides given first are for the primer corresponding to the coding strand, and those given second are for the primer corresponding to the complementary strand. The primers used for the amplification of exons 2B-3, 4-5A, 6-7, and 10-12 correspond, respectively, to nucleotides 1,1961 (22). The oligonucleotides were synthesized on a 381A DNA synthesizer (Applied Biosystems, Foster City, CA).…”

“…A cosmid clone for the human type II collagen gene (COL2A1), designated cosHcol2A (Cheah et al, 1985), was used in the present study. This cosmid clone contains a 36 kb insert.…”
