Identification and Characterization of Terpene Synthases Potentially Involved in the Formation of Volatile Terpenes in Carrot (Daucus carota L.) Roots

Abstract: Plants produce an excess of volatile organic compounds, which are important in determining the quality and nutraceutical properties of fruit and root crops, including the taste and aroma of carrots (Daucus carota L.). A combined chemical, biochemical, and molecular study was conducted to evaluate the differential accumulation of volatile terpenes in a diverse collection of fresh carrots (D. carota L.). Here, we report on a transcriptome-based identification and functional characterization of two carrot terpene… Show more

“…In angiosperms, the TPS-b and TPS-a subfamilies account for most of the sesquiterpene synthases and monoterpene synthases, which constitute the majority of TPS proteins. DCAR_023152 (DcTPS1) and DCAR_012963 (DcTPS2) were two TPSs found in carrot belonging to the TPS-a subfamily and TPS-b subfamily, respectively 67 . The TPSs of celery showed the closest phylogenetic relationships with those of carrot, another Apiaceae family plant.…”

“…Cells were further grown for 12 h at 18°C. Bacteria were lysed and soluble protein purified by nickel chromatography and assayed for purity by SDS-PAGE as previously described by Yahyaa et al (2015). An optimized LnTPS3 ORF for expression in E. coli was synthesized and cloned into pUC57 vector (GenScript).…”

“…High ethanol emissions (1 mg kg -1 ) from carrots and acetaldehyde emissions from carrots (0.5 mg kg -1 ) and zucchini (0.3 mg kg -1 ) could originate from drought stress conditions during the growth, mechanical stress after the harvest or storage under low oxygen conditions 39. Boiling carrots additionally emits considerable amounts of sesquiterpenes (0.3 mg kg -1 ) which are formed during root development in the plants 40.…”
