Identification and Characterization of Choline Transporter-Like Protein 2, an Inner Ear Glycoprotein of 68 and 72 kDa That Is the Target of Antibody-Induced Hearing Loss

Abstract: The Kresge Hearing Research Institute-3 (KHRI-3) antibody binds to a guinea pig inner ear supporting cell antigen (IESCA) and causes hearing loss. To gain insight into the mechanism of antibody-induced hearing loss, we used antibody immunoaffinity purification to isolate the IESCA, which was then sequenced by mass spectroscopy, revealing 10 guinea pig peptides identical to sequences in human choline transporter-like protein 2 (CTL2). Full-length CTL2 cDNA sequenced from guinea pig inner ear has 85.9% identity … Show more

“…The tissue was carefully harvested at the time of labyrinthectomy for Meniere's disease or the labyrinthectomy portion of the translabyrinthine approach for removal of vestibular schwannoma as described previously. 8,12,11 All dissections were performed at room temperature under normal saline. Using an otologic drill, otic capsule bone was removed from the ampullated ends of the semicircular canals.…”

“…In situ hybridization and antisera to cochlin showed cochlin to be expressed in the spiral ligament and limbus , whereas CTL2 is prominently expressed on supporting cells in the organ of Corti (Zajic et al 1991;Ptok et al 1993;Nair et al 2004). Using antibodies raised to unique peptides in each protein, we show in this report that cochlin and CTL2 physically interact and can co-precipitate one another.…”

“…The KHRI-3 monoclonal antibody developed from mice immunized with cell suspensions from the organ of Corti (Zajic et al 1991) binds to the CTL2 protein expressed on inner ear supporting cells (Nair et al 2004). Serum-free KHRI-3 antibody was produced in the Cellmax artificial capillary system (Pierce, Rockford, IL, USA; Nair et al 1997).…”
