Identification and characterization of a new true lipase isolated through metagenomic approach

Abstract: BackgroundMetagenomics, the application of molecular genomics to consortia of non-cultivated microbes, has the potential to have a substantial impact on the search for novel industrial enzymes such as esterases (carboxyl ester hydrolases, EC 3.1.1.1) and lipases (triacylglycerol lipases, EC 3.1.1.3). In the current work, a novel lipase gene was identified from a fosmid metagenomic library constructed with the "prokaryotic-enriched" DNA from a fat-contaminated soil collected from a wastewater treatment plant.Re… Show more

“…This approach also overcomes the problem of cultivation of microbes as majority of the micro organisms are not to cultivation (Handelsman et al, 1998). There were so many recent reports proving the novelty of this approach in the identification of true lipases (Jiang et al, 2006;Lee et al, 2006;Elend et al, 2007;Jeon et al, 2009;Liu et al, 2009;Meilleur et al, 2009;Glogauer et al, 2011).…”

“…In order to carry out the reaction (1 ml), a stock solution of 25 mM p-nitrophenyl butyrate was prepared in acetonitrile-isopropanol (1:4 [vol/vol]) (26) and mixed with 50 mM sodium phosphate buffer (pH 7.0) to obtain a 1 mM substrate final concentration. Control reaction mixtures containing no enzyme were utilized to account for any spontaneous hydrolysis of the substrates tested.…”

“…While a large number of different lipases have been discovered and commercialized, only a few lipases with broad pH, temperature range and high activity against long-chain triglycerides such as vegetable oils have been found so far from the metagenome (Glogauer et al, 2011;Jeon et al, 2009;Jiang et al, 2006). Two soil metagenome-derived lipases, PWTSB and PWTSC, were reported as having an optimum temperature of 20 C and 40 C respectively.…”
